DDIT4 promotes malignancy of head and neck squamous cell carcinoma

Summary: This study investigated the cancer-promoting effect of DDIT4 in Head and Neck Squamous Cell Carcinoma (HNSCC) and its mechanisms. DDIT4 overexpression was found to be associated with worse tumor characteristics, while DDIT4 knockdown inhibited the proliferation, invasion, and migration of HNSCC cells, as well as the expression of HIF-1a, VEGF, and vimentin. Additionally, DDIT4 expression was negatively correlated with immune infiltrations and played a crucial role in the differentiation of CAFs and T cells.

Oligoprogression in MSI-H/dMMR patients can be salvaged with surgical resection and/or local treatment and continuation of pembrolizumab off study. Further clinical studies are warranted to evaluate different MSI-H/dMMR EC subtypes treated with ICIs.

Copper induces cell death by targeting lipoylated TCA cycle proteins

Summary: Copper is an essential cofactor for all organisms, but excessive copper can lead to cell death. We found that copper-dependent cell death mechanism is different from known mechanisms, and it is related to mitochondrial respiration. Copper directly binds to lipoylated components of the tricarboxylic acid cycle, causing protein aggregation and loss of iron-sulfur cluster proteins, ultimately leading to cell death.

Identification of a novel cuproptosis-related gene signature and integrative analyses in patients with lower-grade gliomas

Summary: Cuproptosis is a newly discovered form of programmed cell death in lower-grade gliomas (LGGs), and its prognostic value and correlation with the tumor microenvironment (TME) have not been studied. In this study, we investigated the genetic and transcriptional variation, prognostic value, and expression patterns of cuproptosis-related genes (CRGs) in LGG. We developed a risk model based on the CRG score to predict LGG patients' prognosis and found that a high CRG score was associated with worse survival outcomes. The CRG score was also correlated with TME characteristics and treatment responses, suggesting its potential as a therapeutic target.

Mechanistically, copper accumulation-induced ATP depletion activates AMPK to promote HMGB1 phosphorylation, resulting in increased release. Inhibiting AMPK activation reduces cuproptosis and HMGB1 release. Cuproptotic cells deficient in HMGB1 show reduced production of inflammatory cytokines. Thus, HMGB1 is a key mediator of sterile inflammation initiated by cuproptosis.

Molecular and immunohistochemical evaluation of BAP-1 antibody in bladder cancer and comparison with luminal-basal subtyping

GATA-3 and CK5/6 showed higher expression in non-invasive and muscle-invasive groups respectively, with a negative correlation between GATA-3 and CK5/6 in the latter. Additionally, the expression of BAP-1 did not correlate with GATA-3 and CK5/6 in any of the groups.
