Identification of Natural Compounds as Inhibitors of Pyruvate Kinase M2 for Cancer Treatment

Iqra Sarfraz et al.

Summary: This study identified ellagic acid, silibinin, curcumin, and resveratrol as potential inhibitors of PKM2 through enzymatic-assay-based screening. These compounds showed different inhibitory effects on PKM2 and demonstrated potential as targeted-cancer therapies.

Silibinin induces metabolic crisis in triple-negative breast cancer cells by modulating EGFR-MYC-TXNIP axis: potential therapeutic implications

Mohammad Askandar Iqbal et al.

Summary: The study shows that silibinin disrupts metabolic reprogramming in TNBC cells through modulation of the EGFR-MYC-TXNIP signaling pathway, leading to decreased cell biomass and proliferation. Silibinin inhibits glycolytic metabolism in TNBC, and combination therapy with paclitaxel yields promising results, highlighting the potential of targeting metabolic vulnerabilities in TNBC treatment.

Kaempferol Reverses Aerobic Glycolysis via miR-339-5p-Mediated PKM Alternative Splicing in Colon Cancer Cells

Haili Wu et al.

Summary: The study shows that kaempferol inhibits colon cancer cell proliferation in a dose-dependent manner, delays cell cycle progression, and induces apoptosis. Kaempferol also affects energy metabolism by regulating the miR-339-5p-hnRNPA1/PTBP1-PKM2 axis, leading to reduced glycolysis and tumor growth inhibition in colon cancer.

C-Glucosylation as a tool for the prevention of PAINS-induced membrane dipole potential alterations

Summary: The study demonstrates that polyphenols can modify cell membrane dipole potential, especially in cholesterol-rich domains, which can lead to interference. By incorporating sugars into polyphenols, compounds are generated that no longer exhibit this interfering behavior.

Thymoquinone, as a Novel Therapeutic Candidate of Cancers

Belal Almajali et al.

Summary: Natural product thymoquinone (TQ) from Nigella sativa has shown promising anticancer activities through inhibiting cell proliferation, migration, and invasion in various human cancers. Its efficacy in inducing apoptosis and mechanisms on different cancer types are still not fully understood, but there is ongoing research on its potential as a future anticancer therapy nominee, especially in combination with other anticancer agents using nanotechnology.

Mangifera indica Extracts as Novel PKM2 Inhibitors for Treatment of Triple Negative Breast Cancer

Azhar Rasul et al.

Summary: This study aimed to identify PKM2 inhibitors from natural products for cancer treatment, with mango and silk cotton tree extracts found to inhibit PKM2. Furthermore, the potential anticancer effects of mango extracts on TNBC were demonstrated through experiments on MDA-MB231 cells.
