Divergent Coupling of 2-Aryl-1,3-Indandiones with Alkenes Enabled by Iodine Catalyst

Summary: In this study, we developed an iodine-catalyzed protocol for the cross-coupling of 2-aryl-1,3-indandiones with alkenes. By varying additives and solvents, we achieved divergent reactivity, leading to the selective synthesis of tertiary alkylated ketones and alkenes. This approach features commercially available starting materials, broad substrate scope, and good tolerance towards various functional groups (including halogens, esters, nitriles, and thioethers). Furthermore, we proposed plausible pathways to explain the divergent selectivity through preliminary mechanism studies.

Palladium-Catalyzed [3+2] Annulation of Aromatic Amides with Maleimides through Dual C-H Activation

Summary: A palladium-catalyzed [3 + 2] annulation of substituted aromatic amides with maleimides is described, providing tricyclic heterocyclic molecules in good to moderate yields through weak carbonyl chelation. The reaction proceeds via a dual C-H bond activation, first selectively at the benzylic position and then at the meta position, resulting in a five-membered cyclic ring. The use of external ligand Ac-Gly-OH is crucial for the success of this protocol. A plausible reaction mechanism has been proposed for the [3 + 2] annulation reaction.

1,3-Migrative Ring Expansion of Spiroindolenines to Azepino[3,4-b]indoles

Summary: We serendipitously discovered an unprecedented ring expansion reaction of 2-alkylspiroindolenines to azepinoindoles, mediated by n-tetrabutylammonium fluoride. The starting materials can be easily prepared by the oxidative dearomative spirocyclization of indole derivatives, catalyzed by hypoiodite. Mildly basic conditions and electron-deficient protecting groups for the amines were found to be crucial for promoting chemoselective reactions. Moreover, the ring expansion of aniline-derived spiroindolenines can proceed smoothly under much milder conditions using only a catalytic amount of cesium carbonate.

Radical Cascade Multicomponent Minisci Reactions with Diazo Compounds

Biocatalytic One-Carbon Ring Expansion of Aziridines to Azetidines via a Highly Enantioselective [1,2]-Stevens Rearrangement

Summary: The study reports the enantioselective one-carbon ring expansion of aziridines to form azetidines through engineered carbene transferase enzymes. A variant of cytochrome P450BM3, P411-AzetS, showed unparalleled stereocontrol (99:1 er) in the [1,2]-Stevens rearrangement and could override the inherent reactivity of aziridinium ylides to perform this rearrangement. By controlling the reactive aziridinium ylide intermediates, these evolvable biocatalysts enable a transformation not achievable by other catalyst classes.

Generation of Fused Seven-Membered Polycyclic Systems via Ring Expansion and Application to the Total Synthesis of Sesquiterpenoids
