Mapping genomic loci implicates genes and synaptic biology in schizophrenia

Summary: In this study, a two-stage genome-wide association study was conducted to identify common variants associated with schizophrenia. The results revealed 287 distinct genomic loci and 120 genes likely to be involved in the development of schizophrenia. Polygenic prediction of educational attainment within and between families from genome-wide association analyses in 3 million individuals

Summary: In this study, a genome-wide association study (GWAS) was conducted to investigate the genetic factors associated with educational attainment (EA). The results identified numerous single-nucleotide polymorphisms (SNPs) that are significantly correlated with EA. Furthermore, a polygenic predictor (PGI) was found to explain a substantial portion of the variation in EA and contribute to disease risk prediction. Intriguingly, the correlation between mate-pair PGIs suggests additional assortment mechanisms. However, no significant SNPs were identified in separate analyses of dominance deviations and the X-chromosome.

Multi-ancestry genetic study of type 2 diabetes highlights the power of diverse populations for discovery and translation

Summary: By conducting genome-wide association studies (GWAS) on ancestrally diverse populations, we identified 237 loci associated with type 2 diabetes (T2D) and further fine-mapped these loci. The majority of T2D associations were found to be driven by a single variant. In addition, our study showed that genetic risk scores derived from ancestrally diverse populations can enhance the transferability of T2D prediction across different populations.

RNA editing underlies genetic risk of common inflammatory diseases

Summary: The study reveals that ADAR-mediated RNA editing may be an important mechanism underlying genetic variants associated with common inflammatory diseases, with disease risk variants associated with reduced editing of cellular double-stranded RNA (dsRNA) and induction of interferon responses and inflammation. This suggests that cellular dsRNA editing and sensing could be an underappreciated mechanism of common inflammatory diseases.

Epigenomic and transcriptomic analyses define core cell types, genes and targetable mechanisms for kidney disease

Genetic control of RNA splicing and its distinct role in complex trait variation

Summary: Most genetic variants identified from GWAS in humans are noncoding and play a role in gene regulation. Previous studies have shown links between GWAS signals and eQTLs, but the links to other genetic regulatory mechanisms, such as sQTLs, have not been extensively explored. This study introduces a new sQTL mapping method and identifies distinct cis-sQTLs that are not associated with eQTLs in brain transcriptomic data. By integrating sQTL data into GWAS for brain-related complex traits, the study identifies genes associated with these traits that cannot be discovered using eQTL data alone.

Where Are the Disease-Associated eQTLs?

Summary: Most disease-associated genetic variants, even though located in potentially regulatory regions, do not show detectable effects on gene expression. This could be due to the fact that gene expression has not been examined in the most relevant cell types or conditions for the disease. Current large-scale efforts to study gene expression are limited to human samples obtained opportunistically or postmortem, usually from adults.
