In Vitro and In Vivo Antitumor Assay of Mitochondrially Targeted Fluorescent Half-Sandwich Iridium(III) Pyridine Complexes

Liyan Wang et al.

Summary: This paper investigates the potential value of half-sandwich iridium(III) complexes with favorable luminescence performance in the anticancer field. Ten fluorescent complexes were prepared and showed potential antiproliferative activity, with Ir6 exhibiting the best activity and excellent fluorescence performance. Ir6 followed an energy-dependent cellular uptake mechanism, specifically accumulating in mitochondria and inducing mitochondrial-mediated apoptosis. It also arrested the cell cycle, catalyzed NADH oxidation, reduced the mitochondrial membrane potential, induced an increase in ROS level, and exhibited an oxidation mechanism. In vivo antitumor assay confirmed that Ir6 effectively inhibited tumor growth and was safer than cisplatin.

Anticancer half-sandwich Ir(III) complex and its interaction with various biomolecules and their mixtures - a case study with ascorbic acid

Lukas Masaryk et al.

Summary: The study investigated the interactions of newly developed metallodrugs with various biomolecules, revealing the reduction and oxidation reactions of azo bonds in the presence of certain biologically relevant molecules. The complexation of the metallodrugs played a crucial role in facilitating these redox reactions, shedding light on the mechanisms of action in extracellular and intracellular environments.

Marzieh Miar et al.

Summary: The study investigates the effects of electron-releasing and electron-withdrawing groups at para positions on the molecular structure of synthesized compounds in selected solvents and in the gas phase. Results show that intramolecular interactions between substituents and the electron donor-acceptor centers of the thiazole ring influence the relative stability of the compounds. Charge-transfer possibilities within the molecules are revealed through dipole moment values and energy gaps between occupied and unoccupied molecular orbitals.

Necroptosis-inducing iridium(iii) complexes as regulators of cyclin-dependent kinases

Ruilin Guan et al.

Summary: NecroIr1 and NecroIr2, iridium(iii) complexes, induce necroptosis in cisplatin-resistant lung cancer cells by selectively accumulating in mitochondria and causing oxidative stress. Additionally, they interfere with cell cycle progression, leading to G0/G1 phase arrest. The combined effect of necroptosis induction and cell cycle arrest makes them potential antitumor agents against drug-resistant cancer cells.
