Summary: The endoplasmic reticulum (ER) is responsible for protein and lipid biosynthesis. ER dysfunction, known as ER stress, can lead to protein accumulation and cell death, contributing to metabolic diseases and cancers. The Unfolded Protein Response (UPR) is activated in response to ER stress, but chronic UPR activation can result in apoptosis. Recent evidence suggests that cells undergoing ER stress can release extracellular signals to propagate UPR activation to target tissues in a cell non-autonomous manner. Additionally, studies have shown that the UPR regulates the biosynthesis of bioactive signaling lipids like ceramides. This article proposes that UPR activation during ER stress drives the biosynthesis of ceramide lipids, which function as cell non-autonomous signals to propagate UPR activation.

Do Amino Acid Antiporters Have Asymmetric Substrate Specificity?

Summary: Amino acid antiporters facilitate the exchange of amino acids. Our study using LC-MS showed that different antiporters have varying substrate specificity. Most antiporters are symmetric, equalizing the pools of their substrate amino acids, except for y(+)LAT1 and y(+)LAT2 which co-transport neutral amino acids with Na+ ions. ASCT1 and ASCT2 antiporters selectively transported glycine and proline as influx substrates. These findings suggest that antiporters can exhibit non-canonical modes of transport.

We evaluated the effect of intermediate storage temperature and period on analyte concentrations in plasma samples using LC-MS, and made recommendations for sample-handling protocols. Certain analytes were found to be unstable, emphasizing the need for meticulous processing. These recommendations will improve the reliability and quality of samples for routine clinical diagnosis involving metabolites.
