Viscosity decoupling does not guarantee dynamic heterogeneity: A way out

Summary: This study developed a new analysis method for identifying dynamic heterogeneity in a better way. The analysis selectively probed different diffussion time-regimes in a heterogeneous system from a single measurement and observed their temperature evolution. The temperature dependent translational diffusion of a fluorophore was studied using fluorescence correlation spectroscopy. The results suggest that the difference in the extent of viscosity decoupling for different diffusion time-regimes might be a better criteria to identify dynamic heterogeneity.

Summary: Conformational heterogeneity is a defining characteristic of proteins. This study used red edge excitation shift (REES) to investigate the conformational heterogeneity of multi-domain human serum albumin, and found that the behavior and dynamics of side chains vary depending on the domain being examined. In the unfolded state, the extent of REES becomes unimportant due to the presence of free water and disruption of protein structure. Water molecules play a significant role in providing rigidity to domain-II.

Assessing the compatibility of choline-based deep eutectic solvents for the structural stability and activity of cellulase: Enzyme sustain at high temperature

Summary: As a new generation of 'green solvents', deep eutectic solvents (DESs) show promise as alternative solvents. In this study, three choline-based DESs were synthesized and evaluated for their impact on cellulase stability and activity using spectroscopic techniques. The results showed that ChCl-glycerol and ChCl-ethylene glycol improved the structural stability and activity of cellulase while maintaining its thermal stability, whereas ChCl-lactic acid had a detrimental effect on the enzyme.

However, it does not guarantee medium heterogeneity, as the decoupling may also originate from the deviation of the basic assumption of SED. We developed a method to find a stronger relation between viscosity decoupling (p ? 1) and dynamic heterogeneity in terms of rotational motion, using solvatochromic probe measurements at different excitations or emissions.

Hydration in Deep Eutectic Solvents Induces Non-monotonic Changes in the Conformation and Stability of Proteins

Summary: This study demonstrates that the hydration of deep eutectic solvents (DESs), a type of solvent used for preserving labile biomolecules, has a significant impact on the behavior, conformation, and stability of proteins. The behavior of two proteins, bovine serum albumin and lysozyme, and an antibody, immunoglobulin G, in DESs with different hydration levels was investigated. The results showed that the proteins exhibited a re-entrant behavior with solvent hydration, with a dome-shaped transition observed at very low and high hydration levels. The hydration also influenced protein folding and stability. The study highlights the importance of quantifying the hydration of DESs in understanding protein behavior and provides new insights into protein folding and stability in hydrated DESs.

This review article provides the recent developments and efforts on the technological developments carried out with ILs and DESs for the stabilization and activation of proteins and enzymes. The most studied IL- and DES-based formulations for proteins and enzymes are discussed, along with their molecular mechanisms and interactions that lead to increased stability, emphasizing their main advantages.
