Theory of Protein Charge Transfer: Electron Transfer between Tryptophan Residue and Active Site of Azurin

Summary: This study investigates the process of electron transfer in the conductivity relay of azurin through theoretical analysis and molecular dynamics simulations. It is found that oxidation of tryptophan leads to electrowetting of the residue, resulting in nonparabolic free energy surfaces for electron transfer. The medium dynamics, including Stokes-shift dynamics and the modulation of electron tunneling through the donor-acceptor distance, affect protein electron transfer. The equilibrium donor-acceptor distance is determined by protein-water dynamics and is not affected by the probability of electron tunneling, which puts most intraprotein electron-transfer reactions under dynamical control.

Microgravity environment grown crystal structure information based engineering of direct electron transfer type glucose dehydrogenase

Summary: The structure of the subunit interface of a promising enzyme called heterotrimeric flavin adenine dinucleotide dependent glucose dehydrogenase has been elucidated using protein crystals grown under a space microgravity environment. By introducing inter-subunit disulfide bonds, a mutant form of this enzyme has been developed with higher catalytic activity and direct electron transfer ability. This mutant enzyme exhibits stable activity even at higher temperatures, surpassing the capabilities of the current gold standard enzyme, glucose oxidase.

Summary: The bioelectrocatalytic properties of membrane-bound aldehyde dehydrogenase (AlDH) from Gluconobacter oxydans NBRC12528 were evaluated, showing direct electron transfer (DET)-type activity for acetaldehyde oxidation at various electrodes. The coordination of CN with AlDH at the multi-walled carbon nanotube-modified electrode changed the direction of DET-type bioelectrocatalysis under acidic conditions. These findings were discussed in terms of thermodynamics.
