Long term culture promotes changes to growth, gene expression, and metabolism in CHO cells that are independent of production stability

Summary: The phenotypic stability of Chinese hamster ovary (CHO) cells during long term culture (LTC) is a major challenge in therapeutic protein manufacturing. This study explored the effects of LTC on CHO cells, specifically looking at growth, gene expression, and cell metabolism. The results showed that LTC altered the behavior of CHO cells, leading to a decline in cell viability and changes in gene expression and cell metabolism. These changes were associated with alterations in cell transcriptome and metabolome, and had implications for the production stability of recombinant proteins.

Genomic features of recombinant CHO clones arising from transposon-based and randomized integration

Summary: The use of transposase in cell line development programs has become more popular in recent years. This study characterizes the mechanism of action and the genomic and phenotypic characteristics of clones derived from transposase, as well as their impact on long-term bioproduction. The results show that transposase reduces the cellular pool of transient vector after transfection and expedites stable pool establishment. It also improves recombinant DNA expression and antibody chain balance, resulting in higher titers compared to randomly generated pools.

Shortening the biologics clinical timeline with a novel method for generating stable, high-producing cell pools and clones

Summary: Reducing drug development timelines is crucial for delivering medicines to patients in need efficiently. The use of cell pools instead of clones can potentially shorten development timelines, as demonstrated during the COVID-19 pandemic. Our novel technology, GPEx(R) Lightning, allows for rapid and targeted recombination of transgenes into the Chinese hamster ovary cell line genome, enabling the swift production of high-expressing stable cell pools and clones. These pools and clones exhibit stable titers and glycosylation, demonstrating strong similarities in glycosylation profiles.

A genetic off-target event in a site-specific integration cell line expressing monoclonal antibody has no impact on commercial suitability

Summary: Site-specific integration (SSI) cell line systems have advantages for biotherapeutic development and production, but they can still experience rare off-target events and vector rearrangements. In this study, we developed an SSI cell line for producing an IgG1 monoclonal antibody. While evaluating the suitability of clone C10 for commercial use, we found unexpected genomic rearrangements but determined that they had no impact on the cell line's genetic consistency, phenotypic stability, production process, or product quality. Based on the consistent genetic, phenotypic, and process characteristics, clone C10 was considered suitable as a manufacturing cell line.

Deciphering integration loci of CHO manufacturing cell lines using long read nanopore sequencing

Summary: The complete integration locus of two Chinese hamster ovary (CHO) cell lines was reconstructed using CRISPR/Cas9 target enrichment, nanopore sequencing, and the Canu de novo assembly pipeline. The study revealed that one CHO cell line integrated three near full-length vector copies, while another CHO cell line integrated seven highly fragmented vector copies. Comparison with TLA-seq data showed that TLA-seq missed most of the breakpoints of the vector-vector fusions.

Mitochondrial membrane potential-enriched CHO host: a novel and powerful tool for improving biomanufacturing capability

Summary: In this study, we enriched Chinese hamster ovary (CHO) host cells with high mitochondrial membrane potential (MMP) to enhance protein productivity and biomanufacturing capability. Stable transfectant pools and clonal cell lines expressing difficult-to-express bispecific molecules were generated from the MMP-enriched host. The MMP-enriched host exhibited improved fed-batch productivity, enhanced cell viability, favorable lactate metabolism, and improved cell cloning efficiency. Proteomic analysis and Western blot validation revealed the underlying mechanisms by which high MMP influenced production performance.

Towards maximum acceleration of monoclonal antibody development: Leveraging transposase-mediated cell line generation to enable GMP manufacturing within 3 months using a stable pool

Summary: In recent years, the biopharmaceutical industry has focused on accelerating development timelines to bring innovative therapies to patients more quickly. A promising alternative to the traditional random transgene integration method has been identified as transposase-mediated semi-targeted transgene integration, which allows for rapid generation of high-performing and stable production cell lines. By applying this new strategy, a robust drug substance manufacturing process was developed at an unprecedented pace, paving the way for Phase I clinical trials within six months. Additionally, a clonally derived production cell line was successfully implemented, resulting in a significant increase in monoclonal antibody production within eight months.
