Summary: This study constructed a 3D culture environment using GelMA hydrogels with different amino substitution degrees and found that Piezo1 expression was stimulated by the stiff matrix, leading to improved expression of osteogenic markers. It was also observed that Piezo1 can be activated by the static mechanical conditions of the stiff matrix, promoting intracellular calcium increase and ATP energy metabolism during cellular differentiation. Additionally, the study revealed the activation of the AMPK-ULK1 axis and modulation of autophagy level in the 3D stiff matrix. Overall, this research contributes to the understanding of the interaction mechanisms between biomimetic extracellular matrix biomaterials and cells and provides a theoretical basis for bone regeneration biomaterials design and application.

Summary: Fibrin is a natural polymer with diverse applications in bioengineering and biomedical fields. However, its characteristics, such as shrinkage, degradation, poor mechanical properties, and batch-to-batch variability, limit its broad use. This review critically examines recent advances in composite fibrin scaffolds, chemically modified fibrin hydrogels, and fibrin-based interpenetrated polymer network (IPN) hydrogels for tissue engineering.

Summary: Cardiovascular diseases are a significant burden on global health services, and impaired vascular function is an important preliminary step in their development. Dairy proteins, specifically whey proteins, contain bioactive peptides and amino acids that have been shown to have beneficial effects on vascular function, including reducing blood pressure. This paper reviews the evidence from randomized controlled trials, animal models, and in vitro models on the effects of whey proteins and their peptides and amino acids on vascular function. It concludes that there is increasing evidence supporting the positive impact of whey proteins on various aspects of vascular function and highlights their beneficial effects on blood pressure, arterial stiffness, nitric oxide production, and inflammation.

Summary: Aging is a major risk factor for microvascular dysfunction, but the underlying mechanism is not fully understood. This study demonstrates that changes in the mechanical properties of tissue collagen may contribute to endothelial cell dysfunction. Using a hydrogel model, the researchers found that increasing matrix stiffness led to the disruption of vascular networks. Inhibiting cell contractility prevented this disruption, partially rescuing the age-related vascular phenotype.

Summary: Recent advances in coronary stents have shown that surface modification, such as titania nanotexturing, can enhance re-endothelialization and reduce restenosis. In this study, titania-coated cobalt-chromium stents with nanotopography were found to facilitate preferential cell response, leading to rapid re-endothelialization and minimal neointimal hyperplasia. These drug and polymer-free stents could potentially serve as an alternative to clinical drug-eluting stents.

Summary: This study focused on the design and evaluation of novel hydrogels based on whey protein isolate (WPI) for tissue regeneration. Modified with bioactive glass (BG) and produced through heat-induced gelation, the hydrogels showed high porosity with homogeneous distribution of BG particles, affecting pore size and reducing material porosity. The presence of BG particles improved thermal stability, decreased enzymatic degradation, and promoted mineralization in simulated biological fluids. In vitro tests demonstrated cytocompatibility and support for osteoblastic cell functions in the hydrogels.
