Young Exosome Bio-Nanoparticles Restore Aging-Impaired Tendon Stem/Progenitor Cell Function and Reparative Capacity

MicroRNA expression in extracellular vesicles as a novel blood-based biomarker for Alzheimer's disease

Summary: By analyzing microRNAs in small extracellular vesicles in the blood, unique cellular and molecular information related to the onset and progression of Alzheimer's disease can be obtained. The study found that specific microRNAs in certain subtypes of small extracellular vesicles showed differential expression in individuals with normal cognition, mild cognitive impairment, MCI conversion to AD dementia, and AD dementia, and were correlated with cortical thickness on magnetic resonance imaging. These findings suggest that microRNA analysis in specific extracellular vesicles may serve as a novel blood-based molecular biomarker for Alzheimer's disease.

MSC-derived extracellular vesicles as nanotherapeutics for promoting aged liver regeneration

Summary: Aging impairs liver regeneration after hepatic surgery in the elderly population, and no effective treatment is available for clinical administration. Mesenchymal stem cell-derived extracellular vesicles (MSC-EVs) have shown therapeutic potential in liver diseases. This study found that MSCs can reduce senescence-associated markers and promote liver regeneration in aged mice through the secretion of MSC-EVs. Mechanistically, these effects were partially attributed to inducing mitophagy via the Atg4B pathway. The presence of DEAD-Box Helicase 5 (DDX5) in MSC-EVs was found to interact with E2F1 and activate the expression of Atg4B. These findings highlight the clinical application potential of MSC-EVs in preventing severe complications in aged individuals undergoing liver surgery.

Clinical-grade human umbilical cord-derived mesenchymal stem cells improved skeletal muscle dysfunction in age-associated sarcopenia mice

Summary: With the expansion of the aging population, age-associated sarcopenia (AAS) has become a severe clinical disease of the elderly and a key challenge for healthy aging. Unfortunately, there are currently no approved therapies for treating AAS. In this study, human umbilical cord-derived mesenchymal stem cells (hUC-MSCs) were administered to two mouse models, and it was found that they significantly restored skeletal muscle strength and performance by raising the expression of crucial extracellular matrix proteins, activating satellite cells, enhancing autophagy, and impeding cellular aging. This study provides a novel model for AAS and highlights the potential of hUC-MSCs as a promising strategy for treating AAS and other age-associated muscle diseases.

Neural Stem Cell-Derived Extracellular Vesicles Counteract Insulin Resistance-Induced Senescence of Neurogenic Niche

Summary: The depletion of neural stem and progenitor cells (NSPCs) may contribute to cognitive impairment in age-related diseases, and insulin resistance has been linked to brain dysfunction. In an experimental model of insulin-resistant NSPCs, researchers identified a molecular circuit involving insulin receptor substrate-1 (IRS-1)/Forkhead box O (FoxO) signaling cascade that inhibits the activation of transcription factors FoxO1 and FoxO3a, which regulate cell proliferation and self-renewal. Insulin resistance also increased the expression of cyclin-dependent kinase inhibitor 1 (p21) and accelerated NSPC senescence. However, stimulation of NSPCs with NSPC-derived exosomes (exo-NSPC) rescued IRS-1/FoxO activation and countered the reduced proliferation and senescence of stem cells. Administration of exo-NSPC via the nasal route also restored the balance between proliferating and senescent NSPCs in the hippocampus of mice on a high-fat diet, improving adult hippocampal neurogenesis. These findings provide insights into the metabolic control of NSPC fate and the role of extracellular vesicles in regulating cell fate in the adult neurogenic niche.
