Synthesis and biological evaluation of theranostic Trastuzumab-SN38 conjugate for Near-IR fluorescence imaging and targeted therapy of HER2+breast cancer

Summary: Here, researchers developed a new theranostic antibody drug conjugate (ADC) that combines a HER2-specific antibody (trastuzumab) with a near infrared dye (Cy5) and a bioactive metabolite of irinotecan (SN38). The ADC showed specific accumulation and nanomolar anti-breast cancer activity on HER2-positive cell lines. In in vivo studies, the ADC exhibited good targeting ability for HER2+ tumors and higher anticancer potency compared to trastuzumab alone or a mixture of trastuzumab with SN38. The glutathione-responsive self-immolative disulfide carbamate linker used in this study was proven to be successful and could be applied in targeted anticancer therapy.

Synthesis and Application of Two-Photon Active Fluorescent Rhodol Dyes for Antibody Conjugation and In Vitro Cell Imaging

Summary: A novel family of julolidine-containing fluorescent rhodolsequipped with a wide variety of substituents was synthesized using a versatile two-step process. The synthesized compounds were fully characterized and demonstrated excellent fluorescence properties for microscopy imaging. The best candidate was conjugated to the therapeutic antibody trastuzumab through a copper-free strain-promoted azide-alkyne click reaction. The rhodol-labeled antibody was successfully used for in vitro confocal and two-photon microscopy imaging of Her2+ cells.

The reagent demonstrates similar ease of use as NHS esters but with improved site-directed labeling.

Near-infrared fluorescent probes based on a quinoxaline skeleton for imaging nucleic acids in mitochondria

Summary: In this paper, cationic probes 1a and 1b and a neutral dye 1c were designed and synthesized for the detection of nucleic acids. The probes showed excellent selectivity and an ON-OFF fluorescence response to DNA and RNA with linear correlation between fluorescence intensity and concentration. The probes also demonstrated potential for staining nucleic acids in mitochondria, highlighting their usefulness in low toxicity mitochondrial nucleic acid probes.
