Influence of the exposure concentration of dissolved cadmium on its organotropism, toxicokinetic and fate in Gammarus fossarum

Ophelia Gestin et al.

Summary: This study aimed to investigate the effects of Cd concentration on organotropism, toxico-kinetics, and fate of Cd in gammarids. The results showed that Cd concentration did not significantly modulate the bioconcentration capacity and Cd distribution in different organs. However, Cd accumulation rates in organs (except intestines) were dependent on the exposure concentration. The in vivo management of Cd among organs remained qualitatively unchanged according to exposure concentration. This study highlights the importance of implementing toxico-kinetic approaches to enhance the understanding of the toxicity risk of contaminants in invertebrates.

Dynamic Multiple Reaction Monitoring of amphipod Gammarus fossarum caeca expands molecular information for understanding the impact of contaminants

Maxime Lepretre et al.

Summary: Mass spectrometry in multiple reaction monitoring (MRM) mode enables selective and multiplexed quantification of peptides derived from proteins. This study evaluates the feasibility of using dynamic MRM (dMRM) tools to investigate proteomes in sentinel species and demonstrates their potential for discovering new protein biomarkers. A dMRM assay was developed to capture the functional proteome of a freshwater crustacean and assess the effects of metal exposure. Metal-specific signatures were identified as potential biomarkers for tracking metal levels in freshwater ecosystems. This study highlights the potential of dMRM for identifying and developing biomarkers in sentinel species.

Acclimation and transgenerational plasticity support increased cadmium tolerance in Gammarus populations exposed to natural metal contamination in headwater streams

Aureline Lalouette et al.

Summary: This study investigates the tolerance of the crustacean Gammarus fossarum to cadmium contamination and reveals its characteristics and intergenerational transmission. It is found that the cadmium tolerance of contaminated populations is about two times higher than non-contaminated populations, and this tolerance exists in all age classes of the contaminated populations and can be transmitted to offspring. Additionally, it is observed that the tolerance levels significantly decrease when the organisms are transferred to a cadmium-free environment, and the production of tolerant offspring ceases, confirming the non-genetic transmission of cadmium tolerance. These findings highlight the neglected role of plasticity and non-genetic transmission in the long-term exposure of natural populations to environmental contamination.

Response of biomarkers to metals, hydrocarbons and organochlorine pesticides contamination in crabs (Callinectes ornatus and C. bocourti) from two tropical estuaries (Sao Jose and Sao Marcos bays) of the Maranhao State (northeastern Brazil)

Bruna Duarte Pereira Righi et al.

Summary: The response of biomarkers to chemical contamination in crabs from two tropical estuaries in Maranhao State, northeastern Brazil showed significant spatial, seasonal, and annual variability. The study found higher Zn concentration in crabs from Sao Jose Bay and higher Cd concentration with oxidative damage in crabs from Sao Marcos Bay. Levels of lipid peroxidation in crab hepatopancreas were identified as the most reliable biomarker to distinguish the two bays.

Jabulani Ndaba et al.

Summary: Parasites and their hosts show different physiological responses to environmental changes, and metallothioneins (MTs) can be used as indicators of these responses. This study compared the levels of MTs in the ectoparasite Lamproglena clariae and its host fish Clarias gariepinus and found that the concentrations of MTs were linked with changes in water quality.

Toxicogenomic profiling after sublethal exposure to nerve- and muscle-targeting insecticides reveals cardiac and neuronal developmental effects in zebrafish embryos

Hannes Reinwald et al.

Summary: This study characterized the transcriptomic responses to six insecticides in zebrafish embryos, aiming to identify biomarker candidates for specific modes of action. Abamectin showed the most unique signature related to lipid metabolic processes. Differentially expressed genes after imidacloprid exposure were primarily associated with immune system and inflammation. The findings provide insights into early molecular events in response to the analyzed modes of action.

Organ-specific accumulation of cadmium and zinc in Gammarus fossarum exposed to environmentally relevant metal concentrations*

Ophelia Gestin et al.

Summary: One of the best approaches for improving the assessment of metal toxicity in aquatic organisms is to study their organotropism through kinetic experiments of metal bioaccumulation. This study compared the organ-specific metal accumulation of non-essential (Cd) and essential (Zn) metals in Gammarus fossarum. The results showed different distribution patterns of Cd and Zn concentrations, slower elimination of Cd, and a major role of intestines in the uptake of waterborne Cd and Zn.

Quantification of metallothionein-III in brain tissues using liquid chromatography tandem mass spectrometry

Adam P. Gunn et al.

Summary: A standardized liquid chromatography tandem mass spectrometry method was developed for robust determination of metallothionein-3 (MT3) in human brain tissue extracts, with endopeptidase Lys-C digestion proving to be more accurate and stable than trypsin digestion. The choice of target peptide in the alpha-domain was critical for accurate MT3 detection. The method was demonstrated to be useful in comparing MT3 expression in post-mortem brain tissues of Alzheimer's disease patients and age-matched controls.

One and multi-compartments toxico-kinetic modeling to understand metals' organotropism and fate in Gammarus fossarum

Summary: The study found that G. fossarum efficiently takes up Cd and eliminates it more rapidly compared to the mussel M. galloprovincialis. The organs that accumulate and depurate the most concentrations of both metals are the caeca and intestine. The one-compartment TK model is most relevant for Hg, while the multi-compartments TK model fits Cd data better, demonstrating dynamic transfer of Cd among organs.
