A versatile Halo- and SNAP-tagged BMP/TGFβ receptor library for quantification of cell surface ligand binding

Jerome Jatzlau et al.

Summary: A TGF beta/BMP receptor library with Halo- and SNAP-tags was generated, and a ligand surface binding assay was developed to assess preferential ligand-receptor binding in cells. This assay enables visualization and quantification of receptor-dependent ligand binding using fluorescently labeled ligands. The LSBA can be applied to test different ligands, mutant screens, and evolutionary comparisons, providing insights into ligand-receptor binding dynamics and signaling specificity.

Competition between type I activin and BMP receptors for binding to ACVR2A regulates signaling to distinct Smad pathways

Szabina Szofia Szilagyi et al.

Summary: Activins and bone morphogenetic proteins (BMPs) can compete for common activin type II receptors, providing a novel mechanism to balance signaling between Smad2/3 and Smad1/5/8.

A Baseline for Skeletal Investigations in Medaka (Oryzias latipes): The Effects of Rearing Density on the Postcranial Phenotype

Claudia Di Biagio et al.

Summary: The study investigates the impact of rearing density on skeletal anomalies in medaka. Results show that higher density reduces growth and leads to decreased mineralization of bones and increased incidence of anomalies. Specific changes in the skeletal structure are observed, providing insights into the etiology of anomalies and the molecular mechanisms involved.

Neuron-Radial Glial Cell Communication via BMP/Id1 Signaling Is Key to Long-Term Maintenance of the Regenerative Capacity of the Adult Zebrafish Telencephalon

Gaoqun Zhang et al.

Summary: The study demonstrates the crucial role of BMP and Id1 signaling in maintaining neural stem cells in adult zebrafish brain and how they interact with neurons to regulate NSC pool. Furthermore, BMP/Id1 signaling also cooperates with Notch signaling through the transcriptional regulator her4.1 to affect the proliferation and regeneration of NSCs. These findings highlight a complex regulatory network involved in NSC maintenance and regeneration in adult zebrafish brain.

ActivinA Induced SMAD1/5 Signaling in an iPSC Derived EC Model of Fibrodysplasia Ossificans Progressiva (FOP) Can Be Rescued by the Drug Candidate Saracatinib

Susanne Hildebrandt et al.

Summary: Balanced signal transduction is crucial in tissue patterning, especially in the vasculature. Aberrant ActivinA signaling in FOP endothelial cells leads to differential vascularity in FOP lesions, which can be partially rescued by the drug candidate Saracatinib. This study provides new insights into the vascular differences in FOP lesions.
