Biocatalyzed Synthesis of Glycostructures with Anti-infective Activity

Pilar Hoyos et al.

Summary: Molecules with carbohydrate moieties are important in fighting bacterial and viral infections, making the design of new carbohydrate-containing drugs or vaccines a topic of great interest. Conventional chemical synthesis methods for these compounds face challenges, and the search for sustainable processes using biocatalysts and eco-friendly solvents is vital. Enzymes have become preferred catalysts in glycochemistry and glycobiology, and their use in synthesizing carbohydrate-based drugs has grown rapidly in recent years. This field represents an example of how biocatalysis can contribute to the development of glycan-based therapies and the fight against antibiotic resistance.

Development of sustainable synthesis of glucuronic acid glycodendrimers using ball milling and microwave-assisted CuAAC reaction

Cecilia Garcia-Oliva et al.

Summary: Two green strategies, solvent free mechanochemistry and microwave irradiation using a recycable biosolvent, based on two activation pathways, for copper-catalyzed azide-alkyne cycloaddition reaction were reported. Glucuronic acid glycodendrimers were synthesized with good conversions (99%), with the microwave pathway being the ideal tool (90% yield in 90 min).

Sulfonated and Carboxymethylated β-Glucan Derivatives with Inhibitory Activity against Herpes and Dengue Viruses

Jose Louzinho Lopes et al.

Summary: By introducing carboxyl and sulfonate groups into β-D-glucan, researchers attempted to improve its antiviral activity against enveloped viruses. The results showed that sulfonated-carboxymethylated derivatives could inhibit herpes virus, while their inhibition of dengue virus was weaker, indicating varying resistance of different viruses to anionic β-D-glucans. Comparing with a previous study, the presence of carboxymethyl groups might have a negative impact on antiviral activity.

Design, Synthetic Strategies, and Therapeutic Applications of Heterofunctional Glycodendrimers

Leila Mousavifar et al.

Summary: Glycodendrimers have attracted significant interest in biomedical applications due to the diverse glycan architectures on cell surfaces, impacting cancer cell metastasis, pathogen recognition, immune cell communications, and vaccine development. Since their inception in 1993, the synthesis of glycodendrimers has evolved into sophisticated methodologies. This review highlights common synthetic approaches and modern applications of heterofunctional glycodendrimers in nanomolecular therapeutics and synthetic vaccines.

Cu(I)-Catalyzed Click Chemistry in Glycoscience and Their Diverse Applications

Anand K. Agrahari et al.

Summary: The review discusses the successful utilization of CuAAC or click chemistry in the field of glycoscience, focusing on the synthesis of triazole-appended carbohydrate-containing molecular structures such as glycohybrids, glycoconjugates, glycopolymers, glycopeptides, glycoproteins, glycolipids, glycoclusters, and glycodendrimers through regioselective triazole forming modular and bio-orthogonal coupling protocols. These glycoproducts find widespread applications as enzyme inhibitors in drug discovery and development, sensing, gelation, chelation, glycosylation, and catalysis. Additionally, the review provides insights into the impact of click chemistry and outlines future perspectives in various emerging disciplines of science and technology.
