Fernando Tobias Silveira et al.

Summary: Visceral leishmaniasis (VL), also known as kala-azar, is caused by the Leishmania (L.) infantum chagasi parasite, which has been found to have a high genomic similarity (99%) with L. (L.) infantum. Understanding the characteristics of this disease and its transmission in the New World can aid in the development of targeted prevention and treatment strategies.

Role of Treg, Breg and other cytokine sets in host protection and immunopathology during human leishmaniasis: Are they potential valuable markers in clinical settings and vaccine evaluation?

Francesca Divenuto et al.

Summary: Leishmaniasis is a vector-borne disease caused by protozoan parasites. Cytokines produced by immune cells play crucial roles in the infection progression and host protection. Pro-inflammatory cytokines produced by Th1 cells promote macrophage activation and parasite killing, while high levels of cytokines produced by Th2, Treg, and Breg cells inhibit macrophage activation and facilitate parasite escape. Further investigations are needed to understand the role of Treg and Breg cytokines in diagnosis, prognosis, and vaccine development for leishmaniasis.

The cytokine/chemokine response in Leishmania/HIV infection and co-infection

Semer Maksoud et al.

Summary: HIV infection weakens the immune system and allows opportunistic infections such as leishmaniasis to occur. Both HIV and leishmaniasis infect certain types of cells and can modulate the activity of other cells. This review discusses the cytokine/chemokine response in leishmaniasis and HIV infection and co-infection, analyzing the potential for targeted therapies.

Leishmanicidal Activity of Guanidine Derivatives against Leishmania infantum

Fernanda Silva Almeida et al.

Summary: Leishmaniasis is a neglected tropical infectious disease, particularly visceral leishmaniasis, which is of great concern to global health. Guanidine-bearing compounds showed antimicrobial activity against Leishmania infantum and caused significant apoptosis in amastigotes. Further research is needed to fully understand their mechanism of action and potential as anti-leishmanial molecules.

Slc11a1 gene polymorphism influences dextran sulfate sodium (DSS)-induced colitis in a murine model of acute inflammation

Stephane Tereza Queiroz de Andrade et al.

Summary: Ulcerative Colitis (UC) is an inflammatory disease associated with an increased risk of carcinogenesis. Genetic studies have shown that gene variants of the SLC11A1 protein are associated with UC susceptibility, with mice carrying the Slc11a1 S allele being more susceptible to DSS-induced colitis.

T Lymphocyte Exhaustion During Human and Experimental Visceral Leishmaniasis

Juliana C. Costa-Madeira et al.

Summary: A key aspect of immunity against Leishmania parasites is the development of an optimal T cell response, which can be compromised during chronic infection, leading to T cell exhaustion. Multiple factors, including parasite load, infection duration, and host immunity, contribute to T cell exhaustion. This can result in disease progression and acquired resistance to conventional drugs.

Estimating the global demand curve for a leishmaniasis vaccine: A generalisable approach based on global burden of disease estimates

Sakshi Mohan et al.

Summary: This study aims to provide a method for estimating the commercial and public health value of a vaccine in development by relying on publicly available Global Burden of Disease data. The estimation of demand for a future leishmaniasis vaccine suggests that it could be commercially attractive to potential manufacturers based on the projected demand and ability-to-pay of countries.

Treatment outcome of imported cutaneous leishmaniasis among travelers and migrants infected with Leishmania major and Leishmania tropica: a retrospective study in European centers 2013 to 2019

Hedvig Glans et al.

Summary: This article describes and evaluates the treatment outcomes of cutaneous leishmaniasis (CL) among travelers and migrants in Europe caused by Leishmania major and Leishmania tropica. The study found that L. major infections were mostly found in individuals visiting friends and relatives, while L. tropica infections were mainly identified in migrants. Pentavalent antimony had a cure rate of 73% (L. major) and 78% (L. tropica) among patients.

Distinct plasma chemokines and cytokines signatures in Leishmania guyanensis-infected patients with cutaneous leishmaniasis

Tirza Gabrielle Ramos de Mesquita et al.

Summary: The immunopathology associated with Leishmaniasis is a consequence of inflammation that may lead to different clinical manifestations depending on host-immune response type, with various pathways influencing disease severity. Comparing immune profiles in patients with cutaneous leishmaniasis caused by L. guyanensis and healthy individuals helps identify specific immune factors related to disease development progression. Multiple biomarker pairs showed high correlation, with cytokines IL-17 and IL-1 beta exhibiting the most positive pair correlation among patients. Prediction of disease progression in Lg-infected individuals may be associated with specific cytokines.

Cutaneous and Mucocutaneous Leishmaniasis

I Abadias-Granado et al.

Summary: Leishmaniasis is a chronic disease caused by flagellate protozoa of the genus Leishmania, with the three main types being cutaneous, mucocutaneous, and visceral. Diagnosis is done through direct microscopic examination or molecular analysis, while treatments vary but evidence for options available for cutaneous leishmaniasis is weak. The disease presentation and management have changed in recent decades due to acquired immune deficiency from conditions like HIV infection or TNF inhibitors.

Potential biomarkers of immune protection in human leishmaniasis

Mahmoud Nateghi Rostami et al.

Summary: Leishmaniasis is a vector-borne neglected tropical disease endemic in over 100 countries worldwide, posing a threat to human health. Lack of a comprehensive biomarker as a surrogate marker of protection or exacerbation remains a barrier for vaccine development. Most understanding of the role of different markers of immune response in the disease has been collected from experimental animal models.
