Mitochondria ROS and mitophagy in acute kidney injury

Summary: Mitophagy is a crucial mitochondrial quality control mechanism that removes damaged mitochondria and reactive oxygen species (ROS) production. The relationship between mitochondrial oxidative stress, ROS production, and mitophagy is closely intertwined and involved in various pathological conditions of acute kidney injury (AKI). The complex process of mitophagy in mammals involves multiple pathways and its interaction with different types of cell death, such as apoptosis, pyroptosis, and ferroptosis in kidney injury, remains unclear. This review discusses recent advances in understanding the relationship between ROS and mitophagy, the different pathways of mitophagy, the interplay between mitophagy and cell death, and their significance in the pathogenesis of AKI.

Summary: Acute kidney injury (AKI) is a global public health problem without specific therapy, characterized by excessive inflammation. Phosphoglycerate mutase 5 (PGAM5) has been found to be up-regulated and associated with kidney dysfunction in AKI. Knockout of PGAM5 alleviated kidney injury by reducing mitochondrial abnormality and inflammatory cytokine production. The release of mitochondrial DNA (mtDNA) induced by PGAM5-mediated Bax dephosphorylation and the activation of cGAS-STING pathway are critical determinants of inflammation and kidney injury in AKI.

MiR-20a-5p alleviates kidney ischemia/reperfusion injury by targeting ACSL4-dependent ferroptosis

Summary: Ischemia/reperfusion injury (IRI) often occurs after kidney transplantation, causing delayed graft function (DGF). MicroRNAs, especially miR-20a-5p, play a significant role in the development of ischemia/reperfusion-induced acute kidney injury and DGF. This study discovered that miR-20a-5p is upregulated in acute kidney injury mice and DGF patients' kidneys. The experiment also showed that the mimic of miR-20a-5p could attenuate IRI and renal fibrosis, while the inhibitor of miR-20a-5p aggravated the injuries. Moreover, the study found that miR-20a-5p is a negative regulator of ACSL4, which is involved in ferroptosis, a type of cell death, suggesting a potential therapeutic application in kidney transplantation.

Kidney fibrosis: from mechanisms to therapeutic medicines

Summary: Chronic kidney disease (CKD) affects 10-14% of the global population, and kidney fibrosis is a hallmark manifestation of progressive CKD. However, there are currently no antifibrotic therapies available for CKD. Understanding the complex interplay between injured parenchyma and non-parenchymal cell lineages in the fibrotic niche, as well as the role of genetics and epigenetics, has shed light on the mechanisms of kidney fibrosis. Recent advancements in single-cell technology have helped answer key questions about profibrotic renal tubules, myofibroblast origins, immune cell involvement, and cell communication. Additionally, the reversible nature of epigenetic changes offers potential therapeutic strategies to halt or reverse kidney fibrosis. Developing biomarkers and antifibrotic treatments for CKD are also important areas of research.
