Epoxide Hydrolases: Multipotential Biocatalysts

Marek Bucko et al.

Summary: This review discusses the state of the art and development potential of epoxide hydrolases as biocatalysts, focusing on their application as chiral building blocks for bioactive compounds and drugs. The review covers new approaches to discover epoxide hydrolases using genome mining and enzyme metagenomics, as well as techniques to improve enzyme activity, enantioselectivity, enantioconvergence, and thermostability. The study also explores possibilities for expanding the synthetic capabilities of epoxide hydrolases through non-natural enzyme cascade reactions.

New Trends and Perspectives in Production of 1,2-Propanediol

Marek Glowka et al.

Summary: The production of 1,2-propanediol is facing significant challenges in the chemical industry due to the increasing demand and environmental regulations. The traditional method is energy intensive, therefore sustainable routes using renewable raw materials are becoming more attractive. This perspective reviews the current research and patents on 1,2-propanediol production, discussing alternative pathways, such as glycerol hydrogenolysis and biotechnological methods, and modern methods like chromium-free catalytic systems and multifunctional catalysts. Biobased 1,2-propanediol has the potential to replace traditional petroleum-derived 1,2-propanediol with environmental and economic benefits.

Metabolic engineered E. coli for the production of (R)-1,2-propanediol from biodiesel derived glycerol

Wilson Sierra et al.

Summary: The development of the biodiesel industry has negatively affected the glycerol market as it produces glycerol as a major byproduct. By applying circular economy concepts, progress has been made in using glycerol residue as a starting material. Metabolic engineering in E. coli has resulted in the design of biocatalysts capable of transforming glycerol into (R)-1,2-propanediol with high purity.

Insights into C1 and C3 assimilation pathways in type I methanotrophic bacterium from co-production of 1,2-propanediol and lactate

Hoa Thi Quynh Le et al.

Summary: The study explores the metabolic pathways of methane and methanol to produce 1,2-propanediol (1,2-PDO) using the type I methanotroph Methylotuvimicrobium alcaliphilum 20Z. It finds that the carbon flux from methane to the EMP pathway restricts the production of 1,2-PDO. Co-utilization of C1 substrates and glycerol enhances the production of 1,2-PDO, although lactate is co-produced in higher amounts. The study provides insights into metabolic routes for 1,2-PDO and lactate co-production from C1 and C3 compounds in methanotrophs.
