Innovative strategies for photodynamic therapy against hypoxic tumor

Xiaotong Li et al.

Summary: Photodynamic therapy (PDT) is a powerful therapeutic option for tumors, but the hypoxic tumor microenvironment limits the supply of oxygen and affects the effectiveness of PDT. Innovative strategies to relieve tumor hypoxia and enhance PDT efficiency have been developed, including O2-independent PDT and synergistic therapies. Future studies should focus on addressing the challenges and exploring the prospects of these strategies.

High-Coverage Four-Dimensional Data-Independent Acquisition Proteomics and Phosphoproteomics Enabled by Deep Learning-Driven Multidimensional Predictions

Moran Chen et al.

Summary: 4D DIA-based proteomics is a promising technology, but it is limited by the time-consuming building of a project-specific experimental library. In this study, a versatile deep learning model called Deep4D was developed to predict multiple parameters for both unmodified and phosphorylated peptides, enabling high-coverage 4D DIA proteomics and phosphoproteomics workflows based on multidimensional predictions. A 4D predicted library containing millions of peptides was established, allowing experimental library-free DIA analysis and identifying more proteins compared to a single-shot measurement with an experimental library in the example of HeLa cells.

Photodynamic therapy: Innovative approaches for antibacterial and anticancer treatments

Marketa Kolarikova et al.

Summary: Photodynamic therapy is an alternative treatment for cancer and bacterial infections that was discovered in 1900 by Oscar Raab and Professor Hermann von Tappeiner. It involves the administration of a photosensitizer followed by light irradiation to induce reactive oxygen species and cell death. While this treatment is not new, there are ongoing modifications and improvements in terms of the photosensitizer's selectivity and efficiency, as well as the types of light sources used.

Spotlight on porphyrins: Classifications, mechanisms and medical applications

Fuyu Yang et al.

Summary: Photodynamic therapy (PDT) and sonodynamic therapy (SDT) are non-invasive treatment methods that effectively inhibit tumors with minimal side effects. Sensitizers play a crucial role in determining the therapeutic effect of PDT and SDT. Porphyrins, a group of organic compounds widely found in nature, can be activated by light or ultrasound to generate reactive oxygen species. Therefore, porphyrins have been extensively studied as sensitizers for PDT and SDT. This review summarizes the applications and mechanisms of classical porphyrin compounds in PDT and SDT, and also discusses their potential in clinical diagnosis and imaging.

Dithiocarbazate-FeIII,-CoIII,-NiII, and-ZnIIComplexes: Design,Synthesis, Structure, and Anticancer Evaluation

Yi Gou et al.

Summary: Non-platinum-metal complexes have shown great potential as anticancer drugs. In this study, a series of dithiocarbazate non-Pt-metal complexes were designed and evaluated for their efficacy as antineoplastic agents. Complex 2 exhibited higher anticancer efficacy than other complexes and cisplatin, and showed comparable hemolysis with cisplatin. In vivo studies demonstrated that complex 2 effectively inhibited tumor xenograft growth with low systemic toxicity. Further investigations revealed that complex 2 suppressed cancer cells by simultaneously inhibiting mitochondria and glycolytic bioenergetics.

Multifunctional Nanoplatforms as a Novel Effective Approach in Photodynamic Therapy and Chemotherapy, to Overcome Multidrug Resistance in Cancer

Martin Majernik et al.

Summary: Photodynamic therapy (PDT) for cancer has advantages over conventional therapeutic approaches, but multidrug resistance is a major limitation. The use of dynamic nanoplatforms can minimize the effect of multidrug resistance and significantly enhance the therapeutic efficacy of PDT. Additionally, nanoparticles can induce synergistic effects when used in combination treatments and improve the target potential in multidrug resistant and cancer stem cells.
