A functional corona around extracellular vesicles enhances angiogenesis, skin regeneration and immunomodulation

Martin Wolf et al.

Summary: Nanoparticles derived from therapy-grade human placental-expanded stromal cells are surrounded by a protein corona, which improves skin regeneration and angiogenesis and induces differential signaling in immune cells.

Lipid-based strategies used to identify extracellular vesicles in flow cytometry can be confounded by lipoproteins: Evaluations of annexin V, lactadherin, and detergent lysis

Jaco Botha et al.

Summary: Flow cytometry is commonly used to characterize extracellular vesicles (EVs). However, this study found that PS labeling and detergent lysis methods may be confounded by lipoproteins. It suggests using transmembrane proteins to differentiate EVs from lipoproteins.

Human milk extracellular vesicles target nodes in interconnected signalling pathways that enhance oral epithelial barrier function and dampen immune responses

Marijke Zonneveld et al.

Summary: Maternal milk extracellular vesicles play a crucial role in supporting epithelial barrier function and modulating the immune system. Their unique proteome can target key hotspots of signaling networks in various cell types within the gastrointestinal tract.

Formation of a protein corona on the surface of extracellular vesicles in blood plasma

Eszter A. Toth et al.

Summary: This study found that a protein corona is formed around EVs in blood plasma, with shared EV corona proteins identified. These results help explain the commonly reported plasma protein 'contamination' of EV preparations and offer a new perspective for EV research.

Robust sequential biophysical fractionation of blood plasma to study variations in the biomolecular landscape of systemically circulating extracellular vesicles across clinical conditions

Glenn Vergauwen et al.

Summary: This study fractionated blood plasma using SEC and OptiPrep density gradient centrifugation to investigate clinical and time-dependent variations in the biomolecular landscape of systemically circulating EV. Results showed that EV carry context-dependent and/or time-dependent protein and small RNA compositions, with differential analysis providing a catalogue of proteins not associated with EV. Overall, blood plasma fractionation can advance the biological understanding and biomarker development of systemically circulating EV.
