A polygenic risk score for Alzheimer's disease constructed using APOE-region variants has stronger association than APOE alleles with mild cognitive impairment in Hispanic/Latino adults in the US

Tamar Sofer et al.

Summary: This study used polygenic risk scores (PRSs) to investigate the association between Alzheimer's disease (AD) risk and mild cognitive impairment (MCI) in a sample of Hispanic/Latino older adults. The results showed that a sum of AD PRSs was associated with MCI, and the association was stronger when including the APOE gene region. This suggests that variants in the APOE gene other than the classic alleles may be important predictors of MCI in Hispanic/Latino adults.

The Brain-Derived Neurotrophic Factor Functional Polymorphism and Hand Grip Strength Impact the Association between Brain-Derived Neurotrophic Factor Levels and Cognition in Older Adults in the United States

Tingting Liu et al.

Summary: Aging is associated with subtle cognitive decline, and lower BDNF levels have been linked to cognitive decline in older adults. This study found that the association between serum BDNF levels and cognitive function may vary based on the BDNF Val66Met genotype. The association also differs according to hand grip strength, but not physical activity.

Association between polygenic risk for Alzheimer's disease, brain structure and cognitive abilities in UK Biobank

Summary: This study investigated the association between LOAD-PGR and structural brain MRI and cognition in healthy adults. Results showed that LOAD-PGR was related to declines in fluid intelligence, matrix completion, smaller left hippocampal volumes, and aspects of cognitive ability. No significant interactions were observed between APOE status and LOAD-PGR in fully adjusted models.

Summary: This study estimated the prevalence of Mild Cognitive Impairment (MCI) in Type 2 Diabetes Mellitus (T2DM) patients through systematic review and meta-analysis. The results showed a high prevalence of MCI in T2DM patients worldwide, especially in China and Asia. Female patients had a higher prevalence of MCI compared to male patients, and older patients had a lower prevalence compared to younger patients.

Polygenic Risk Score for Low-Density Lipoprotein Cholesterol Is Associated With Risk of Ischemic Heart Disease and Enriches for Individuals With Familial Hypercholesterolemia

Haoyu Wu et al.

Summary: The study suggests that an LDL-C PRS can be utilized to identify individuals with FH variants, and the association between ischemic heart disease and the LDL-C PRS is similar to that of measured LDL-C, likely due to the PRS reflecting lifetime exposure to LDL-C levels.

Identifying individuals with high risk of Alzheimer's disease using polygenic risk scores

Ganna Leonenko et al.

Summary: Polygenic Risk Scores for Alzheimer's disease offer unique possibilities for reliably identifying individuals at high and low risk. The study shows that the best prediction accuracy is achieved with a model including APOE and a polygenic score excluding APOE. Standardizing PRS against the population mean makes individuals' scores comparable between studies.

Polygenic risk scores for Alzheimer's disease are related to dementia risk in APOE e4 negatives

Jenna Najar et al.

Summary: This study found an association between AD-PRS and dementia, particularly in APOE e4 non-carriers. APOE e4 was associated with an increased risk of dementia, and in individuals aged 95 years and older, APOE e4 only predicted dementia in the low-risk tertile of AD-PRS.

Summary: Alzheimer's disease has a significant impact on individuals and society as a whole, with a large number of patients in the United States and a potential for continued growth in the future. The value of unpaid caregivers has increased despite a decline in their numbers, and healthcare costs are on the rise. Racial and ethnic disparities in healthcare persist, highlighting the need for greater diversity in dementia care workforce and clinical trials.

Summary: The study found that APOE ε4 is associated with an increased risk of cognitive decline in Latinos, with varying effects across different Latino backgrounds, and Amerindian genetic ancestry can protect against the risk conferred by APOE ε4 on cognitive decline.
