Identifying Balanced Chromosomal Translocations in Human Embryos by Oxford Nanopore Sequencing and Breakpoints Region Analysis

Zhenle Pei et al.

Summary: In this study, a clinically applicable method was developed to reduce the risk of pregnancy loss for reciprocal translocation carriers. Long-read Nanopore sequencing and breakpoint analysis accurately diagnosed chromosomal reciprocal translocations in embryos. This method is important for selecting embryos with a normal karyotype.

Intragenic inversions in NF1 gene as pathogenic mechanism in neurofibromatosis type 1

Viola Alesi et al.

Summary: This study reports two NF1 patients with different inversion mutations that disrupt the coding sequence of the NF1 gene and result in the NF1 phenotype. Some patients who cannot obtain a molecular diagnosis using current methods may have undetectable pathogenic variants due to the limitations of current technologies.

Marfan Syndrome Caused by Disruption of the FBN1 Gene due to A Reciprocal Chromosome Translocation

Anna Clara Schnause et al.

Summary: This study reports a mother and daughter with marfanoid habitus, both showing a cytogenetically reciprocal balanced translocation between chromosomes 2 and 15 after no pathogenic variant in the FBN1 gene was found. The breakpoints in the FBN1 gene were identified through various methods, highlighting the importance of structural genome variants as an underlying cause of monogenic diseases.

Clinical genetic diagnostics in Danish autosomal dominant polycystic kidney disease patients reveal possible founder variants

Marlene L. Nielsen et al.

Summary: A study on Danish patients with ADPKD found that 87% of patients had pathogenic or possibly pathogenic variants, with a total of 112 observed variants, 94 of which were unique. Recurrent variants were observed in 26% of families, with some likely being founder variants.

Optical genome mapping enables constitutional chromosomal aberration detection

Tuomo Mantere et al.

Summary: The study investigated the use of optical genome mapping (OGM) to detect known chromosomal aberrations, with results showing OGM's ability to accurately detect various types of chromosomal abnormalities, including complex rearrangements. High-resolution OGM achieved 100% concordance compared to standard assays for aberrations without centromeric breakpoints.
