Regulating tumor cholesterol microenvironment to enhance photoimmunotherapy in oral squamous cell carcinoma

Chuanhui Song et al.

Summary: Immunotherapy is hindered by the immune suppression caused by high cholesterol concentration in the tumor microenvironment. To address this issue, researchers have developed a cholesterol-regulate nanoplatform that can reduce cholesterol levels and activate the immune system to enhance the effectiveness of tumor immunotherapy. Both in vitro and in vivo experiments have demonstrated that this nanoplatform can effectively kill tumor cells, regulate cholesterol levels, and stimulate the immune system, providing a new strategy for overcoming immune suppression in the tumor microenvironment and paving the way for adjuvant immunotherapy.

Self-oxygenated co-assembled biomimetic nanoplatform for enhanced photodynamic therapy in hypoxic tumor

Bingchen Zhang et al.

Summary: Photodynamic therapy (PDT) has great potential in cancer treatment, and its important effect in inhibiting tumors is the activation of immunogenic cell death (ICD) effects. However, the strategy is limited in the innate hypoxic tumor microenvironment. This study developed self-oxygenated biomimetic nanoparticles (CS@M NPs) to overcome this limitation and achieve enhanced PDT in triple negative breast cancer (TNBC) treatment. The results demonstrated that CS@M NPs effectively activated the immune system and achieved nearly 90% tumor suppression rate in orthotopic TNBC models.

Endotoxin tolerance induced by Porphyromonas gingivalis lipopolysaccharide alters macrophage polarization

Jian-Yu Gu et al.

Summary: Endotoxin tolerance induced by P. gingivalis LPS resulted in a reduced inflammatory response and enhanced wound healing abilities in macrophages. The tolerant macrophages showed an intermediate state between M1/M2 polarization, functioning as M2-like cells. The PTP1B-MEK1/2-STAT6 signaling pathway may play a role in the polarization of tolerant macrophages.

Summary: This study presents the concept of a next-generation sonodynamic sensitizer preparation for immunosonodynamic therapy (iSDT) as a noninvasive synergistic therapeutic modality. By combining sonodynamic therapy and immunotherapy, this method achieves efficient and targeted treatment for tumors. The iSDT specifically activates the sonodynamic effect at the tumor site, showing high specificity in killing tumor cells. Additionally, when combined with an alpha-PD-L1 antibody, this therapy effectively inhibits tumor metastasis.

Porphyromonas gingivalis Virulence Factors and Clinical Significance in Periodontal Disease and Coronary Artery Diseases

Lorena Horvat Aleksijevic et al.

Summary: Porphyromonas gingivalis is a pathogenic bacterium that can cause periodontal disease and cardiovascular problems. It can enter the bloodstream through periodontal lesions and spread to other organs, which can lead to the development of subacute endocarditis and cardiovascular diseases. P. gingivalis has several virulence and pathogenicity factors, including fimbriae, hemolysin, hemagglutinin, capsule, etc. Additionally, P. gingivalis in biofilm form is less sensitive to antimicrobial drugs, posing a challenge in treating infections caused by this bacterium.

NUPR1 promotes the proliferation and metastasis of oral squamous cell carcinoma cells by activating TFE3-dependent autophagy

Tengfei Fan et al.

Summary: Oral squamous cell carcinoma (OSCC) is the most common type of oral malignancy, and metastasis is the main cause of poor prognosis. This study identifies nuclear protein 1 (NUPR1) as the most significantly upregulated protein in OSCC tumor samples, and high expression of NUPR1 is associated with OSCC tissue and low survival rates. The study also shows that NUPR1 maintains autophagic flux and lysosomal functions by increasing transcription factor E3 (TFE3) activity, thereby promoting OSCC cell proliferation and metastasis in vitro and in vivo.
