NOX Dependent ROS Generation and Cell Metabolism

Tiziana Pecchillo Cimmino et al.

Summary: Reactive oxygen species (ROS) are highly reactive molecules that can induce cytotoxicity or regulate cellular physiology. Among the ROS, the superoxide anion radical (O2 center dot-) plays a key role in redox signaling, generated by the NADPH oxidase (NOX) enzyme family and the mitochondrial electron transport chain. In this review, the authors discuss the emerging roles of NOX enzymes in modulating cancer cell metabolism and highlight their potential as therapeutic targets for rewiring tumor metabolism.

Evaluation of Tamarix nilotica Fractions in Combating Candida albicans Infections

Ehssan Moglad et al.

Summary: The antifungal properties of Tamarix nilotica fractions against Candida albicans clinical isolates were evaluated. The dichloromethane (DCM) and ethyl acetate (EtOAc) fractions showed strong antifungal activity and reduced the biofilm formation capacity of the treated isolates. In a mouse infection model, the DCM fraction decreased the bacterial count in lung tissues and maintained the lung tissue architecture. Furthermore, the DCM fraction significantly reduced the expression of inflammatory cytokines in the lung tissues.

Summary: Metal nanoparticles show great potential as antibacterial agents to combat antibiotic resistance. In this study, zinc oxide nanoparticles (ZnO-NPs) were synthesized using Gardenia thailandica methanol extract (GTME) and their antibacterial activity against Pseudomonas aeruginosa clinical isolates was investigated in vitro and in vivo. The ZnO-NPs exhibited significant antibacterial activity with a decrease in membrane integrity and an increase in membrane permeability. Furthermore, the ZnO-NPs protected lung, liver, and kidney tissues from the deleterious effects of P. aeruginosa bacteria.

Characterization of the Anti-Biofilm and Anti-Quorum Sensing Activities of the β-Adrenoreceptor Antagonist Atenolol against Gram-Negative Bacterial Pathogens

Simona Cavalu et al.

Summary: The development of bacterial resistance to antibiotics is a growing public health concern exacerbated by the formation of biofilms. The study aimed to evaluate the anti-quorum sensing (QS), anti-biofilm, and anti-virulence activities of the beta-adrenoreceptor blocker atenolol against Gram-negative bacteria. In silico studies showed that atenolol had a significant binding affinity to the QS receptors of the tested bacteria. In vitro and in vivo experiments demonstrated that atenolol effectively competed with QS proteins, downregulated the expression of QS- and virulence-encoding genes, and reduced bacterial biofilm formation, virulence enzyme production, and motility. It also protected mice from bacterial infection. These findings suggest that atenolol has potential as an adjuvant in the treatment of aggressive bacterial infections.

Disrupting quorum sensing alters social interactions in Chromobacterium violaceum

Sonia Mion et al.

Summary: Quorum sensing is a communication system used by bacteria to coordinate biological functions in a cell density-dependent manner. Chromobacterium violaceum utilizes acyl-homoserine lactone-based QS to regulate production of antimicrobial and toxic compounds, with targeting AHLs leading to significant changes in bacterial interactions. These findings highlight the key role of AHL-based QS in C. violaceum's interactions with microorganisms and the potential impact of quorum quenching on microbial population dynamics.
