Rebecca L. Siegel et al.

Summary: Each year, the American Cancer Society compiles data on cancer occurrence and outcomes in the United States. The latest data shows that breast and prostate cancer progress has stagnated, while lung cancer has shown improvements in survival rates. Lung cancer incidence for advanced disease has declined while localized-stage rates have increased, resulting in higher survival rates. Mortality patterns align with incidence trends, with lung cancer deaths declining rapidly, breast cancer deaths slowing, and prostate cancer deaths stabilizing.

The discovAIR project: a roadmap towards the Human Lung Cell Atlas

Malte D. Luecken et al.

Summary: The Human Cell Atlas consortium aims to establish an atlas of all organs in the healthy human body at single-cell resolution to increase our understanding of biological processes and accelerate disease diagnosis and treatment. The Lung Biological Network of the HCA aims to generate the Human Lung Cell Atlas as a reference for normal lung homeostasis. The discovAIR project aims to create the first draft of an integrated Human Lung Cell Atlas, which will be a valuable resource for researchers, clinicians, and patients.

Single Cell RNA Sequencing Identifies a Unique Inflammatory Macrophage Subset as a Druggable Target for Alleviating Acute Kidney Injury

Weijian Yao et al.

Summary: This study reveals the complementary roles of kidney resident macrophages (KRMs) and monocyte-derived infiltrated macrophages (IMs) in modulating tissue inflammation and promoting tissue repair in ischemia reperfusion injury induced acute kidney injury (IRI-AKI). S100a9(hi)Ly6c(hi) IMs are identified as early responders to AKI, mediating the initiation and amplification of kidney inflammation. Targeting S100a8/a9 signaling with small-molecule inhibitors demonstrates renal protective effects.

Prognostic value of cartilage intermediate layer protein 1 in chronic heart failure

Can Wang et al.

Summary: The study found that circulating CILP-1 is an independent prognostic predictor in chronic HF, with higher levels associated with increased risk of mortality. Additionally, CILP-1 improved prognostic prediction over N-terminal pro-brain natriuretic peptide, indicating its potential value in predicting patient outcomes.

Osteopontin/secreted phosphoprotein-1 harnesses glial-, immune-, and neuronal cell ligand-receptor interactions to sense and regulate acute and chronic neuroinflammation

Ashley Yim et al.

Summary: Osteopontin (OPN), also known as secreted phosphoprotein-1 (SPP1), is a multifunctional protein expressed in various cell types and plays important roles in intercellular communication and extracellular matrix. OPN possesses cytokine, chemokine, and signal transduction functions by interacting with integrins and CD44-variant receptors. In the brain, OPN is involved in early detection of inflammation and tissue damage.

CILP, a Putative Gene Associated With Immune Infiltration in Breast Cancer Brain Metastases

Xiaolin Sun et al.

Summary: This study found that breast cancer brain metastases (BCBM) are closely related to the expression of cartilage intermediate layer protein (CILP). Low expression of CILP is associated with poor prognosis and is an independent prognostic factor for stage III-IV breast cancer. CILP expression is downregulated in BCBM and negatively correlated with vascular endothelial growth factor A (VEGFA). CILP inhibits breast cancer cell proliferation and metastasis by participating in epithelial-mesenchymal transition (EMT). CILP is also associated with immune infiltration in BCBM, possibly through involvement in the T-cell receptor signaling pathway.

Estimation of tumor cell total mRNA expression in 15 cancer types predicts disease progression

Shaolong Cao et al.

Summary: This study presents a method to quantify tumor-specific total mRNA expression from bulk sequencing data and validates it in various cancer types. High total mRNA expression is associated with increased risk of disease progression and death. The expression is influenced by cancer-specific gene alteration, intra-tumor genetic heterogeneity, and pan-cancer trends in metabolic dysregulation.

Single-cell analyses highlight the proinflammatory contribution of C1q-high monocytes to Behcet's disease

Wenjie Zheng et al.

Summary: This study provides insights into the immune landscape of Behcet's disease (BD) and highlights the contribution of C1q-high (C1q(hi)) monocytes to BD hyperinflammation. C1q(hi) monocytes demonstrate increased phagocytosis and proinflammatory cytokine secretion, and their presence is clinically relevant. Activated interferon-gamma (IFN-gamma) signaling induces the accumulation of C1q(hi) monocytes, which can be reduced by tofacitinib treatment.

SPP1 is a prognostic related biomarker and correlated with tumor-infiltrating immune cells in ovarian cancer

Wen Gao et al.

Summary: This study found that SPP1 is overexpressed in ovarian tumors and high expression is associated with poor prognosis. Additionally, SPP1 expression is positively correlated with infiltrating levels of multiple immune cells, suggesting that SPP1 may play an important role in immune regulation in ovarian cancer.
