LPS-induced vein endothelial cell injury and acute lung injury have Btk and Orai 1 to regulate SOC-mediated calcium influx

Xiaochen Qiu et al.

Summary: This study found that endothelial cell damage in sepsis patients is related to calcium overload, mediated by Orai 1 and TRPC 1. Btk plays a crucial role in regulating calcium overload in LPS-induced endothelial cells, with knockdown inhibiting the expression of relevant channels and cell damage.

Endothelial TRPV4 channels prevent tumor growth and metastasis via modulation of tumor angiogenesis and vascular integrity

Anantha K. Kanugula et al.

Summary: TRPV4 plays a critical role in tumor growth, angiogenesis, and metastasis, with endothelial TRPV4 being a key modulator of vascular integrity and tumor angiogenesis. Deletion of TRPV4 promotes tumor angiogenesis, growth, and metastasis.

Tumor-Derived Extracellular Vesicles Induce Abnormal Angiogenesis via TRPV4 Downregulation and Subsequent Activation of YAP and VEGFR2

Brianna Guarino et al.

Summary: This study demonstrates that t-EVs induce endothelial cell phenotypic transition and abnormal angiogenesis through post-translational downregulation of TRPV4 and activation of the Rho/Rho kinase/YAP/VEGFR2 pathways. Furthermore, t-EVs can induce abnormal vessel formation in vivo independently of tumors, suggesting their potential as novel targets for vascular normalization and cancer therapy.

STIM1 Regulates Endothelial Calcium Overload and Cytokine Upregulation During Sepsis

Xiaochen Qiu et al.

Summary: STIM1-mediated store-operated Ca2+ entry plays a crucial role in calcium overload and inflammation in vascular endothelial cells during systemic inflammatory response and sepsis. Knockdown of STIM1 inhibits inflammatory cytokine expression induced by sepsis serum or LPS, while overexpression of STIM1 promotes induction of these cytokines. Depletion of STIM1 or the SOC proteins TRPC1 and ORAI1 significantly inhibits calcium influx and expression of inflammatory cytokines induced by septic serum or LPS.
