Co-Crystalline Solid Solution Affords a High-Soluble and Fast- Absorbing Form of Praziquantel

Summary: In this study, the solid form characteristics of the solid solutions of praziquantel with both enantiomers of malic acid and tartaric acid were investigated. Two new cocrystals were discovered and three non-stoichiometric, mixed crystal forms were identified and isolated. Thermal and solubility analysis showed that the newly prepared solid solutions had a fourfold solubility advantage over the pure drug. In addition, a pharmacokinetic study in rats was conducted, and innovative mini-capsules were used for the oral administration of the solid samples. The available data suggested that the faster dissolution rate of the solid solutions resulted in faster absorption of the drug and helped maintain a constant steady-state concentration.

Trends in oral small-molecule drug discovery and product development based on product launches before and after the Rule of Five

Summary: This review analyzes the trends in oral drug product development between 1994 and 1997 and between 2013 and 2019. The analysis confirms that most new oral drugs adhere to the 'Rule of Five' (Ro5) limitations, but there is a slight increase in the number of drugs with molecular weight (MW) and calculated partition coefficient (clogP) beyond the Ro5. The observed trend is likely due to incremental advances in various fields such as molecular biology, drug design, and drug delivery.

Switching polymorph stabilities with impurities provides a thermodynamic route to benzamide form III

Summary: The study demonstrates that benzamide form I can easily convert into benzamide form III in the presence of nicotinamide using mechanochemistry. This transformation is due to a thermodynamic switch between these two polymorphic forms driven by the formation of solid solutions with small amounts of nicotinamide. The presence of nicotinamide promotes the robust and exclusive crystallisation of the elusive form III of benzamide.
