Post-Translation Modifications and Mutations of Human Linker Histone Subtypes: Their Manifestation in Disease

Ashok Kumar et al.

Summary: Linker histones play a critical role in chromatin structure and various cellular processes. Recent advancements in mass spectrometry have allowed the identification of post-translational modifications (PTMs) on linker histone subtypes and their potential relevance in diseases. Whole-genome sequencing studies have also revealed mutations in linker histones associated with diseases. In this review, the PTMs of linker histone subtypes, their relevance in human diseases, and the correlation with mutations are compiled.

Structural basis of RNA polymerase II transcription on the chromatosome containing linker histone H1

Rina Hirano et al.

Summary: The authors used cryo-EM to investigate the mechanism of how RNA polymerase II (RNAPII) transcribes DNA in the presence of linker histone H1. They revealed that H1 binding restricts the orientation of the linker DNA and that the transcription elongation factor Spt4/5 reduces H1-mediated RNAPII pausing.

Summary: This study used cryo-EM to analyze the structures of chromatosomes containing different H1 isoforms, revealing that the C-terminal tails of the H1 isoforms mainly control the orientations of flanking DNA. The results provide insights into the structures and dynamics of chromatosomes, with implications for the structure and function of chromatin.

Interactome of Site-Specifically Acetylated Linker Histone H1

Eva Hoellmueller et al.

Summary: Linker histone H1 plays a key role in chromatin organization and maintenance. Using site-specifically mono- and di-acetylated linker histone H1.2 generated by genetic code expansion, this study identified and characterized the acetylation-dependent cellular interactome of H1.2, demonstrating a role in modulating protein-protein interactions. Among the interacting partners identified are translational initiation factors and transcriptional regulators.
