Summary: This study investigated the role of MdASMT9 in modulating melatonin biosynthesis and improving water-use efficiency (WUE) in apple. Overexpression of MdASMT9 led to increased melatonin accumulation and enhanced WUE in transgenic apple lines. Under well-watered conditions, the increased WUE was attributed to an enhancement of photosynthetic rate and stomatal aperture. However, under long-term moderate water deficit conditions, regulations in photoprotective mechanisms, stomatal behavior, osmotic adjustment, and antioxidant activity contributed to improved WUE in transgenic apple lines.
