The role of autophagy in viral infections

Tong Chen et al.

Summary: Autophagy is a cellular process that degrades virus particles and components during a viral infection. However, viruses have evolved various strategies to subvert autophagy and counteract cellular antiviral responses.

The prion protein and its ligands: Insights into structure-function relationships

Mohsin Shafiq et al.

Summary: The prion protein is a multifunctional protein that exists in different folding states. It undergoes various proteolytic processing steps, resulting in the production of membrane-bound and soluble prion protein fragments. In addition to proteinaceous binding partners, the prion protein can interact with metal ions and nucleic acids. However, there is limited information on the structural and functional consequences of the prion protein binding to its partners. This review discusses the structure-function relationship of the prion protein and its binding partners, taking into account the different folding states and fragments.

Nafamostat mesylate as a broad-spectrum candidate for the treatment of flavivirus infections by targeting envelope proteins

Yunzheng Yan et al.

Summary: This study found that Nafamostat (NM) has the potential to inhibit flavivirus infections. Through in vitro and in vivo experiments, researchers found that NM effectively inhibits the replication of Zika virus and other flaviviruses. Further research revealed the antiviral mechanism of NM and proposed a novel drug design approach targeting the flavivirus envelope protein.

Investigating SH-SY5Y Neuroblastoma Cell Surfaceome as a Model for Neuronal-Targeted Novel Therapeutic Modalities

Pooja Gangras et al.

Summary: This study identifies high-confidence surface proteins expressed on the brain and DRG neurons using a bioinformatic workflow and demonstrates significant overlap between the SH-SY5Y cell surfaceome and the surface proteome of human brain and DRG neurons. It also finds that alternative splicing of specific isoforms in 32% of shared surface proteins allows them to maintain their protein-coding ability and localize to the cell surface. Validation of one of these isoforms confirms its presence in neuroblastomas, making it a receptor of interest for neuroblastoma-specific therapeutics.

The SH-SY5Y human neuroblastoma cell line, a relevant in vitro cell model for investigating neurotoxicology in human: Focus on organic pollutants

Lorena Lopez-Suarez et al.

Summary: Investigating the toxicity of chemicals on the human brain traditionally relies on animal and cell models, which have limitations due to species differences, ethical concerns, and time/cost constraints. Human cell models, such as the SH-SY5Y cell line, offer a relevant alternative for studying neurotoxicity and prioritizing in vivo testing. This review examines the characteristics and use of the SH-SY5Y cell line for studying the effects of organic environmental pollutants on neurotoxicity, highlighting the impacts on cellular processes and neurotransmitter homeostasis.

Japanese encephalitis virus manipulates lysosomes membrane for RNA replication and utilizes autophagy components for intracellular growth

Qiang Xu et al.

Summary: The Japanese encephalitis virus manipulates cellular secretory pathways and autophagy components to create an optimal environment for genome replication. The virus replicates in lysosomes, not autophagosomes, and utilizes autophagic components for intracellular growth, shedding light on potential antiviral targets.

Pathobiology of Japanese encephalitis virus infection

Kiran Bala Sharma et al.

Summary: Japanese encephalitis virus (JEV) is a flavivirus transmitted by Culex mosquitoes, causing Japanese encephalitis (JE) which has a high mortality rate and potential for global spread. The disease can result in death or permanent neurological sequelae, with prognosis influenced by virus-host interactions. The pathogenesis of JEV involves a combination of direct virus-induced neuronal cell death and uncontrolled neuroinflammatory response, with cellular stress responses playing a key role in the disease progression.
