Identification and Characterization of the Determinants of Copper Resistance in the Acidophilic Fungus Acidomyces richmondensis MEY-1 Using the CRISPR/Cas9 System

Summary: This study applied a CRISPR/Cas9 system to the acidophilic fungus Acidomyces richmondensis MEY-1 and identified the determinants of Cu resistance. The Cu-binding transcription factor ArAceA and the Cu-transporting P-type ATPase ArYgA were found to play roles in Cu tolerance and pigmentation, respectively. These findings provide insights into the molecular basis of Cu tolerance in A. richmondensis MEY-1 and highlight the potential of the CRISPR/Cas9 system for studying adaptability mechanisms in acidophilic fungi.

AA16 Oxidoreductases Boost Cellulose-Active AA9 Lytic Polysaccharide Monooxygenases from Myceliophthora thermophila

Summary: In this study, two new fungal oxidoreductases from the AA16 family were characterized. These enzymes were found to be capable of producing H2O2 by oxidizing low-molecular-weight reductants. The interplay between AA16 proteins and MtLPMO9 proteins significantly enhanced cellulose degradation. These findings provide new insights into the functions of copper-dependent enzymes and the interplay of oxidative enzymes within fungal systems to degrade lignocellulose.

Dissecting key residues of a C4-dicarboxylic acid transporter to accelerate malate export in Myceliophthora

Summary: In this study, the mechanism and preference of the C4-dicarboxylic acid transporter AoMAE in Myceliophthora thermophila were investigated. Key amino acid residues, including Arg78 and residue at position 100, were found to play crucial roles in substrate transport and protein stabilization.The mutation T100S significantly increased malate production by 68%. By introducing the variant with mutation T100S and deubiquitination, malic acid production in M. thermophila was dramatically increased from 1.22 to 54.88 g/L. These findings enhance our understanding of organic acid transporters and have implications for engineering efficient dicarboxylic acid transporters.

Heterologous Expression and Biochemical Characterization of a Thermostable Endoglucanase (MtEG5-1) from Myceliophthora thermophila

Summary: In this study, endoglucanase (MtEG5-1) from Myceliophthora thermophila was successfully overexpressed in Pichia pastoris GS115. The purified MtEG5-1 showed optimal activity at pH 5 and 70 degrees C, with good thermal stability. The enzyme demonstrated a molecular weight of 55 kDa and had Km and Vmax values of approximately 6.11 mg/mL and 91.74 mu mol/min/mg at 70 degrees C (pH 5.0), respectively. The activity of MtEG5-1 was enhanced by Mn2+ and inhibited by K+. These findings suggest that MtEG5-1 expressed by P. pastoris GS115 is more heat-tolerant and suitable for future academic research and industrial applications.

The putative methyltransferase LaeA regulates mycelium growth and cellulase production in Myceliophthora thermophila

Summary: This study investigated the role of the putative methyltransferase LaeA in regulating fungal growth and cellulase production. Deletion of laeA in Myceliophthora thermophila resulted in enhanced mycelium growth and glucose consumption. Summary: This study reveals that the l-arabinose transporter MtLat-1 is associated with the repression of hemicellulase gene expression in Myceliophthora thermophila. The absence of MtLat-1 results in decreased l-arabinose uptake and consumption rates, but increased mycelium growth, protein production, and hemicellulolytic activities. Comparative transcriptomic analysis shows a different expression profile in the Delta Mtlat-1 strain, indicating that MtLat-1 is involved in the repression of hemicellulase genes.

Summary: The thermophilic fungus Myceliophthora thermophila secretes various enzymes for cellulose degradation, including cellulases GH5_5, GH6, GH7, and lytic polysaccharide monooxygenases (LPMOs) AA9. The core cellulolytic enzymes were identified as MtCel7A and MtCel5A, and their hydrolytic efficiencies were improved by C1/C4 oxidizing LPMO9H but inhibited by C1 oxidizing LPMO9E and C4 oxidizing LPMO9J. These findings suggest the potential application of C1/C4 oxidizing LPMOs for efficient conversion of cellulose into biofuels and biochemicals.
