Loss of sister kinetochore co-orientation and peri-centromeric cohesin protection after meiosis I depends on cleavage of centromeric REC8

Sugako Ogushi et al.

Summary: Protection of near-centromeric REC8 cohesin and sister kinetochore co-orientation depend on spindle-chromosome complexes (SCCs) in meiosis I, while being lost in meiosis II, resulting in sister chromatid disjunction and production of haploid gametes. The catalytic activity of Separase is crucial for converting kinetochores to a bi-oriented state and deprotecting cohesin in meiosis I.Selective cleavage of REC8 near kinetochores is sufficient to disrupt co-orientation in univalent chromosomes, but may require resolution of chiasmata in bivalents.

Deprotection of centromeric cohesin at meiosis II requires APC/C activity but not kinetochore tension

Valentina Mengoli et al.

Summary: This study explores the mechanisms of genome haploidization involving cohesin protection and deprotection at centromeres during meiotic divisions. It is found that APC/C activation can lead to sister centromere separation without bipolar spindle forces in yeast and mouse oocytes. Additionally, the stability of centromeric cohesin at metaphase II requires both Rec8 protection by shugoshin and separase inhibition by securin in yeast. These findings suggest that similar mechanisms preserve chromosome integrity during metaphase II in yeast and prolonged metaphase II arrest in mammalian oocytes.

A highly conserved pocket on PP2A-B56 is required for hSgo1 binding and cohesion protection during mitosis

Yumi Ueki et al.

Summary: The study reveals the structure and function of the PP2A-B56-hSgo1 complex during mitosis, highlighting the essential role of a conserved pocket on the B56 regulatory subunit for hSgo1 binding and cohesion protection. Additionally, it shows that hSgo1 inhibits the binding of PP2A-B56 substrates and that PP2A-B56 dephosphorylates Cdk1 sites on hSgo1 to regulate cohesin interactions. Overall, the research provides important insights into cohesion protection during mitosis.

Borealin directs recruitment of the CPC to oocyte chromosomes and movement to the microtubules

Lin-Ing Wang et al.

Summary: This study investigated the assembly mechanism of the chromosome passenger complex (CPC) in Drosophila oocytes, revealing that the interaction between Borealin and chromatin is crucial for recruiting CPC to chromosomes and playing a key role in spindle assembly. The co-localization of HP1 with CPC on chromosomes promotes the movement of CPC from chromosomes to microtubules, which is essential for spindle assembly and chromosome bi-orientation.

Summary: The study demonstrates that overexpression of E2F3 and Shugoshin-1 in breast cancer promotes tumor metastasis, while their silencing suppresses cell invasion and migration. Additionally, E2F3 and Shugoshin-1 regulate the expression of genes associated with epithelial-to-mesenchymal transition.

Shugoshin ensures maintenance of the spindle assembly checkpoint response and efficient spindle disassembly

Aakanksha Sane et al.

Summary: Shugoshin proteins are evolutionarily conserved and play various roles in maintaining genomic stability in different cells, including regulating chromosome positioning, maintaining chromosomal separation efficiency, and serving as a component of the spindle assembly checkpoint to ensure cell stability. This study in Candida albicans reveals novel functions of Shugoshin, such as facilitating timely disassembly of the mitotic spindle and prolonging the response to improper kinetochore-microtubule attachments.
