Behavior Problems and Social Competence in Fragile X Syndrome: A Systematic Review

Summary: This systematic review examined studies conducted in the past 20 years on behavior and social problems in Fragile X syndrome (FXS), taking into account environmental and personal variables that may influence these problems. The review found that attention deficit hyperactivity disorder (ADHD) remains the most significant behavior problem in FXS, and behavior problems and social competence have remained stable over the past 20 years. Comorbidity with autism in FXS individuals increases behavior problems and worsens social competence profiles. Environmental factors and parental characteristics play a role in behavior problems and social competence.

Multiple Mild Stimulations Reduce Membrane Distribution of CX3CR1 Promoted by Annexin a1 in Microglia to Attenuate Excessive Dendritic Spine Pruning and Cognitive Deficits Caused by a Transient Ischemic Attack in Mice

Summary: This study confirms the impairment of cognition and hippocampal synapses in mice treated with transient ischemic attack (TIA) and demonstrates that this can be reversed by multiple mild stimulations (MMS). The researchers found that TIA promotes the interaction between Annexin a1 (ANXA1) and CX3CR1, leading to increased membrane distribution of CX3CR1 in microglia and enhanced CX3CR1 and CX3CL1 interaction. These effects induced by TIA could be reversed by MMS. Moreover, the study shows that overexpression of ANXA1 in microglia leads to increased membrane distribution of CX3CR1 and CX3CR1-CX3CL1 interaction, as well as reduced spine density in co-cultured microglia and neurons. ANXA1 overexpression in microglia also abolishes the protective effects of MMS after TIA. This study provides a potential strategy for treating delayed synaptic injury caused by TIA.

A short period of early life oxytocin treatment rescues social behavior dysfunction via suppression of hippocampal hyperactivity in male mice

Summary: Early sensory experiences and genes interact to shape precise neural circuits, which is crucial for proper brain function in adulthood. This study found that early life whisker trimming in mice affects social discrimination in adulthood, likely due to enhanced activation of the hippocampal dorsal CA3. Reduced hippocampal oxytocin levels after whisker trimming were also observed, and compensating for oxytocin levels prevented social dysfunction. Similarly, oxytocin treatment was able to recover social behavior deficits in a fragile X syndrome mouse model.
