Genetic and Transcriptional Regulatory Mechanisms of Lipase Activity in the Plant Pathogenic Fungus Fusarium graminearum

These findings provide significant insights into the genetic and regulatory mechanisms underlying lipases in fungi.

Summary: Necrosis and Ethylene-inducing peptide 1-like proteins (NLPs) are found in various bacteria, fungi, and oomycetes. The study investigated the evolutionary history of NLPs in different fungal species and found that the NLP1 family has the strongest cytotoxic activities. The biased retention of NLP1 genes may have contributed to the enhanced cytotoxic effects of this subfamily.

Gene deletion and constitutive expression of the pectate lyase gene 1 (MoPL1) lead to diminished virulence of Magnaporthe oryzae

Summary: This study examined the role of pectate lyases (Pls) in the infection process of Magnaporthe oryzae, the causal agent of rice blast disease. The results showed that the expression of MoPL1 gene significantly increased during the infection process, and deletion or constitutive expression of MoPL1 led to a significant reduction in virulence. Furthermore, it was found that the enzymatic activity of MoPL1 was responsible for the diminished virulence.

It also serves as an excellent plant material for biological research. A comprehensive database of pitaya has been constructed, providing genomic, gene expression, miRNA, metabolite, and proteomic data from different pitaya cultivars. The database also includes videos on flowering process and planting tutorials. These valuable data will significantly facilitate future studies on pitaya genetics, breeding, and function research.

Identification of RT-qPCR reference genes suitable for gene function studies in the pitaya canker disease pathogen Neoscytalidium dimidiatum

Summary: In this study, we evaluated the suitability of eight candidate reference genes for normalizing gene expression levels estimated by RT-qPCR in N. dimidiatum. Tubulin and Actin (1) were identified as the most stably expressed reference genes under different temperatures, while Actin (1) and Actin were the most stably expressed reference genes at different developmental stages. Tubulin and UBQ were the most stably expressed reference genes during interaction with pitaya. Actin and 18s rRNA were the most stably expressed across all experimental conditions. Our results provide insights that will aid future RT-qPCR studies of gene expression in N. dimidiatum.

Comparative proteomic analysis of the fungal pathogen Neoscytalidium dimidiatum infection in the pitaya

Summary: This study used quantitative proteomics to investigate pitaya immune responses against the pathogenic fungus Neoscytalidium dimidiatum (Nd). The results revealed the involvement of many essential proteins associated with photosynthesis, phytohormone activity, ROS homeostasis, and pathogenic defense responses in the defense against Nd infection. Accumulation of phytohormones auxin and abscisic acid, as well as changes in ROS metabolism, were shown to be important for defense against Nd invasion.

The abundance of secreted hydrolytic enzymes and secondary metabolite biosynthetic gene clusters in the genomes of Botryosphaeria, Macrophomina, Lasiodiplodia, and Neofusicoccum suggest similarities to necrotrophic plant pathogens and endophytes of woody plants.
