Autism spectrum disorder and GABA levels in children with succinic semialdehyde dehydrogenase deficiency

Itay Tokatly Latzer et al.

Summary: This study aims to investigate the relationship between succinic semialdehyde dehydrogenase deficiency (SSADHD) and autism spectrum disorder (ASD), which is related to the dysregulation of gamma-aminobutyric acid (GABA) and the imbalance of excitatory and inhibitory neurotransmission. The results show that the severity of ASD in SSADHD increases with age and is inversely correlated with plasma GABA and gamma-hydroxybutyrate levels. A discriminative analysis reveals that age older than 7 years 2 months and plasma GABA levels less than 2.47 mu M are the threshold values for increased likelihood of ASD in individuals with SSADHD. These findings provide insights into the pathophysiology of ASD and may contribute to early diagnosis and intervention in individuals with SSADHD.

The presence and severity of epilepsy coincide with reduced γ-aminobutyrate and cortical excitatory markers in succinic semialdehyde dehydrogenase deficiency

Itay Tokatly Latzer et al.

Summary: This study investigated the characteristics of epilepsy in succinic semialdehyde dehydrogenase deficiency (SSADHD) and its correlation with GABA-related metabolites and neurophysiologic markers. The findings demonstrated a high prevalence of epilepsy in SSADHD, with its onset and severity associated with age-related decline in GABA levels and cortical inhibition.

Establishment and validation of a clinical severity scoring system for succinic semialdehyde dehydrogenase deficiency

Itay Tokatly Latzer et al.

Summary: This study developed a clinical severity scoring (CSS) system for evaluating the severity of Succinic semialdehyde dehydrogenase deficiency (SSADHD), an inherited metabolic disorder. The CSS consists of five domains and was validated using an objective severity scoring (OSS) system. The results show a significant correlation between the CSS and OSS scores, indicating that the CSS is a reliable tool for clinical use.

Identification of novel deep intronic PAH gene variants in patients diagnosed with phenylketonuria

Xiaohua Jin et al.

Summary: The study identified deep intronic variants in the PAH gene that may impact the genetic diagnosis of PKU, with in silico predictions suggesting potential splice site alterations. Furthermore, experimental analysis confirmed the effects of these variants on pseudo-exon inclusion in PAH mRNA, enhancing current understanding of PAH genotypes and the molecular mechanisms underlying PKU.

Understanding the Molecular Mechanisms of Succinic Semialdehyde Dehydrogenase Deficiency (SSADHD): Towards the Development of SSADH-Targeted Medicine

Henry H. C. Lee et al.

Summary: Succinic semialdehyde dehydrogenase deficiency (SSADHD) is a rare genetic disorder that leads to incomplete metabolism of the neurotransmitter GABA, resulting in various clinical manifestations. This article reviews relevant molecular mechanisms and discusses the challenges in SSADHD gene and enzyme replacement therapies. It also introduces a mouse model that restores SSADH activity.

Compound Heterozygosis in AADC Deficiency and Its Complex Phenotype in Terms of AADC Protein Population

Giovanni Bisello et al.

Summary: Aromatic amino acid decarboxylase (AADC) deficiency is a rare monogenic disease caused by mutations in the ddc gene. It is characterized by severe motor impairments and developmental delay. The number of patients and identified variants has increased in recent years, possibly due to ongoing gene therapy trials. Efforts are being made to achieve early diagnosis and predict clinical outcome, but no clear correlation between genotype and phenotype has been found so far.

Assessing Prevalence and Carrier Frequency of Succinic Semialdehyde Dehydrogenase Deficiency

Kirt Martin et al.

Summary: Pathogenic variants in the ALDH5A1 gene lead to SSADH deficiency, with an estimated global prevalence of 1/460,000 and highest carrier frequencies in East Asian and South Asian populations. However, the pan-ethnic carrier frequency for SSADH deficiency may not be fully represented in gnomAD, requiring further analysis to assess the prevalence of this ultra-rare disease.

Enzyme Replacement Therapy for Succinic Semialdehyde Dehydrogenase Deficiency: Relevance in γ-Aminobutyric Acid Plasticity

Henry Hing Cheong Lee et al.

Summary: SSADHD is a rare metabolic disorder caused by functional impairment of the SSADH enzyme, leading to accumulation of GABA and GHB in the brain, resulting in various neurological symptoms. Research is focusing on safely and feasibly restoring SSADH enzyme through enzyme replacement therapy or gene therapy to treat SSADHD patients.
