Atg1 phosphorylation is activated by AMPK and indispensable for autophagy induction in insects

Haigang Zhao et al.

Summary: Phosphorylation is an important post-translational modification in regulating autophagy in insects. By studying the silkworm, researchers found that BmAtg1c mRNA expression, protein expression, phosphorylation, and autophagy all peaked during larval-pupal metamorphosis. They also identified two phosphorylation sites in BmAtg1c that were activated by BmAMPK, and these sites were required for autophagy induction. The corresponding phosphorylation sites in fruit flies were also found to be functionally conserved for autophagy induction.

20-Hydroxyecdysone counteracts insulin to promote programmed cell death by modifying phosphoglycerate kinase 1

Xin-Le Kang et al.

Summary: The regulation of glycolysis and autophagy during feeding and metamorphosis in holometabolous insects is complex and not fully understood. Insulin and 20-hydroxyecdysone (20E) coordinate the balance between cell proliferation and programmed cell death (PCD) through the posttranslational modification of phosphoglycerate kinase 1 (PGK1). Insulin promotes glycolysis and cell proliferation via PGK1 phosphorylation, while 20E dephosphorylates PGK1 to repress glycolysis and initiate PCD.

Cholesterol derivatives induce dephosphorylation of the histone deacetylases Rpd3/HDAC1 to upregulate autophagy

Wenmei Wu et al.

Summary: HDACs play a crucial role in global gene expression and physiological events. This study revealed that autophagy in silkworm requires BmRpd3, which is regulated by 20E signaling negatively and translocated by MTOR inhibition, inducing autophagy. Cholesterol derivatives, especially hydroxylated cholesterol, facilitate autophagy by dephosphorylating and shuttling BmRpd3/HsHDAC1 in nucleo-cytoplasmic localization via MTOR signaling inhibition.

Analysis of Drosophila Atg8 proteins reveals multiple lipidation-independent roles

Andras Jipa et al.

Summary: Research shows that lipidated Atg8a is essential for autophagy, while its non-lipidated form plays a crucial role in larval midgut elimination and viability. In contrast, Atg8b is expressed only in the male germline, and its loss leads to male sterility without affecting autophagy. High expression of non-lipidated Atg8b in the male germline is necessary for fertility.

Post-translational regulation of autophagy is involved in intra-microbiome suppression of fungal pathogens

Jing Wang et al.

Summary: In this study, antagonistic interactions between the fungus Fusarium graminearum and bacterium Streptomyces hygroscopicus were explored at the molecular level. Streptomyces altered the fungal acetylome, leading to induction of fungal autophagy, with the mechanism involving rapamycin secretion inhibiting TOR and degrading Gcn5, ultimately reducing Atg8 acetylation and promoting autophagy in F. graminearum. This novel post-translational regulation of autophagy initiated by a bacterial antibiotic provides insights into microbial homeostasis in healthy plant microbiomes and offers new possibilities for controlling pathogens.

Summary: The guidelines presented in this article aim to assist researchers in selecting and interpreting methods to examine autophagy, as well as to help reviewers provide constructive critiques of reports focused on these processes. It emphasizes that no single assay is perfect for every situation, and multiple techniques may be needed to properly monitor autophagy in different experimental settings. The article also highlights the importance of targeting multiple autophagy-related genes in genetic approaches to block autophagy effectively.
