Paeoniflorin promotes intestinal stem cell-mediated epithelial regeneration and repair via PI3K-AKT-mTOR signalling in ulcerative colitis

Yujing Ma et al.

Summary: Ulcerative colitis (UC) is a chronic and immune-mediated inflammatory disorder. Paeoniflorin (PF), extracted from Paeonia lactiflora, has anti-inflammatory and immunoregulatory efficacy. This study investigated how PF regulates intestinal stem cells (ISCs) to improve the regeneration and repair of the intestinal epithelium in UC. PF significantly alleviated colitis and ameliorated intestinal mucosal injury by regulating ISCs through the PI3K-AKT-mTOR signaling pathway. PF treatment may be beneficial to mucosal healing in patients.

Is the C-reactive protein-to-albumin ratio the most remarkable simple inflammatory marker showing active disease in patients with axial spondyloarthritis, psoriatic arthritis, and rheumatoid arthritis?

Huseyin Kaplan et al.

Summary: The objective of this study was to determine which of the simple inflammation markers obtained from routine laboratory tests showed active disease best and to determine their cutoff value. The results showed that the C-reactive protein to albumin ratio (CAR) was the most important marker to distinguish active disease in patients with axial spondyloarthritis, psoriatic arthritis, and rheumatoid arthritis.

Identification, isolation and analysis of human gut-associated lymphoid tissues

Peter B. Jorgensen et al.

Summary: The authors present a protocol for isolating and studying human gut-associated lymphoid tissue, which allows identification and isolation of all human GALT and GALT-free intestinal lamina propria. This technique involves mechanical separation of intestinal mucosa from the submucosa to identify and isolate specific types of lymphoid tissue for downstream cellular and molecular characterization. The protocol, taking 4-10 hours, could be useful for researchers interested in intestinal immune development and function in health and disease.

Analysis of Antidepressant Activity of Huang-Lian Jie-Du Decoction Through Network Pharmacology and Metabolomics

Shu-Yue Qu et al.

Summary: Depressive disorder is a prevalent mental disorder characterized by depressed mood and lack of interest or pleasure. Herbal medicine, such as Huang-lian Jie-du Decoction (HLJDD), has been explored as a complementary therapy for depression. This study demonstrates that HLJDD exhibits antidepressant effects by modulating tryptophan metabolism, serotonergic and dopaminergic synaptic activities, cAMP signaling pathway, and calcium signaling pathway, suggesting potential targets for its antidepressant action.

Total glucosides of Paeony restores intestinal barrier function through inhibiting Lyn/Snail signaling pathway in colitis mice

Xin-Yue Cao et al.

Summary: The study found that TGP effectively improves symptoms and pathological changes, reduces levels of inflammatory cytokines in the colon tissues and serum in a dose-dependent manner, and restores intestinal barrier function by regulating tight junction and adherens junction proteins. Additionally, TGP inhibits Lyn/Snail signaling pathway and maintains tight and adherens junctions, indicating its potential as a therapeutic agent for IBD.

Adenosine deaminase modulates metabolic remodeling and orchestrates joint destruction in rheumatoid arthritis

Sai Krishna Srimadh Bhagavatham et al.

Summary: The study shows that different levels of adenosine deaminase (ADA), an arthritis-associated factor, can lead to varying cytokine levels and metabolic characteristics in rheumatoid arthritis (RA) patients. ADA may play a crucial role in the pathophysiology of RA joints, serving as a potential biomarker and therapeutic target for RA patients.

Summary: Mitochondrial dysfunction and oxidative stress are associated with T cell-mediated autoimmune diseases, and targeting metabolic pathways can inhibit autoreactive T cell activation. Increasing the requirements of Tregs for ROS and oxidative phosphorylation can promote self-tolerance and inhibit the activity of autoreactive T cells.
