Internal RNA 2′O-methylation in the HIV-1 genome counteracts ISG20 nuclease-mediated antiviral effect

Priscila El Kazzi et al.

Summary: This study reveals that the 2' O-methylation marks inside the HIV-1 genome help the virus evade the host immune system. The virus achieves self-methylation by recruiting the cellular methyltransferase FTSJ3, which allows it to avoid detection by the antiviral protein ISG20. This mechanism enables HIV-1 to replicate and spread more effectively.

Interplay of RNA 2'-O-methylations with viral replication

Alice Decombe et al.

Summary: Viral RNAs (vRNAs) can be modified by methylation, which affects the biological functions of RNA related to sequence, structure, and protein interactions. 2'-O-methylation was initially found to target the cap structure of mRNA, and recently, it was also reported in vRNA. The consequences of these methylations are still unclear, as they have been associated with both pro-viral and anti-viral effects. This article focuses on the mechanisms and potential roles of vRNA 2'-O-methylation in viral replication.

MODOMICS: a database of RNA modification pathways. 2021 update

Pietro Boccaletto et al.

Summary: MODOMICS database, established since 2006, has evolved from containing basic chemical structures of modified ribonucleosides to a comprehensive resource with new functionalities such as a catalog of RNA modifications linked to human diseases. The database, which is extensively linked to RCSB Protein Data Bank, provides users with an enhanced tool for research on RNA modification.

Quantitative profiling of pseudouridylation dynamics in native RNAs with nanopore sequencing

Oguzhan Begik et al.

Summary: This study demonstrated the potential of nanopore RNA sequencing for detecting and identifying various RNA modifications, including pseudouridine and 2'-O-methylation. By focusing on pseudouridine modification sites, the researchers revealed the distribution and dynamics of these modifications in cellular RNAs. The development of a software tool to estimate modification stoichiometries at each site provides a valuable tool for further research on RNA modifications.

Comprehensive mapping of SARS-CoV-2 interactions in vivo reveals functional virus-host interactions

Siwy Ling Yang et al.

Summary: The study identified functional structural elements within the SARS-CoV-2 genome, showed differences in RNA folding between wildtype and mutant viruses, and found hundreds of RNA-RNA interactions within the virus genome and between virus and host RNAs. The virus genome strongly binds to mitochondrial and small nucleolar RNAs and undergoes extensive 2'-O-methylation.

pre-piRNA trimming and 2′-O-methylation protect piRNAs from 3′ tailing and degradation in C. elegans

Benjamin Pastore et al.

Summary: The study found that the 3' termini of Caenorhabditis elegans piRNAs undergo nontemplated nucleotide addition, leading to extensive base-pairing interaction with their target RNAs. Both PARN-1 and HENN-1 enzymes collaboratively protect piRNAs from tailing and degradation through pre-piRNA trimming and 2'-O-methylation.
