An Oxidative Enzyme Boosting Mechanical and Optical Performance of Densified Wood Films

Summary: Nature has developed enzymes that can alter the wood cell wall structure, providing eco-friendly ways to customize the microstructure of wood for high-performance materials. In this study, an oxidative enzyme called lytic polysaccharide monooxygenase (LPMO) was used to modify the cell wall structure of delignified wood under mild reaction conditions. The enzymatic oxidation led to nanofibrillation of cellulose microfibril bundles, allowing the delignified wood to be densified into transparent anisotropic films under ambient conditions and low pressure. The enzymatic nanofibrillation promoted microfibril fusion and improved the adhesion between neighboring wood fiber cells, resulting in enhanced mechanical performance of the films in both longitudinal and transverse directions. This research advances our understanding of LPMO-induced microstructural changes in wood and provides an environmentally friendly alternative to harsh chemical treatments and energy-intensive densification processes, thus contributing to sustainable production of high-performance wood-derived materials.

Designing liquefaction and saccharification processes of highly concentrated starch slurry: Challenges and recent advances

Increasing the initial concentration of starch slurry can reduce the production cost, but it presents challenges such as low reaction efficiency and poor product performance. This study reviews the effects of substrate concentration on starch sugar production and proposes several regulation strategies like pretreatment, enzyme-assisted methods, and temperature control to overcome these challenges. The future of highly concentrated starch sugar production looks promising, and further research is needed to understand the structure and enzyme attack mode for targeted product production.

Expression and characterization of a novel lytic polysaccharide monooxygenase, PdLPMO9A, from the edible fungus Pleurotus djamor and its synergistic interactions with cellulase in corn straw biomass saccharification

Summary: A novel gene, PdLPMO9A, was found to be upregulated during the degradation of corn straw. Heterologous expression of PdLPMO9A showed superior thermostability and pH stability. PdLPMO9A significantly improved the cellulase-mediated saccharification of corn straw, and addition of CuSO4 further increased glucose yield.

Synergistic effects of multiple enzymes from industrial Aspergillus niger strain O1 on starch saccharification

Summary: This study identified key enzymes involved in the starch saccharification process, including glucoamylase, alpha-amylase, and acid alpha-amylase, out of 29 glycoside hydrolases from the 6-day fermentation products of A. niger O1. Synergistic effects were observed among the components of glucoamylases cocktail, with an increase in alpha-amylase leading to improved efficiency of starch saccharification. Overexpression of acid alpha-amylase in strain O1 resulted in an increase in total glucoamylase activity.
