Host defense peptide mimicking antimicrobial amino acid polymers and beyond: Design, synthesis and biomedical applications

Summary: Microbial infections pose a threat to human health and life. Conventional antibiotics are challenged by the emergence of antibiotic-resistant pathogens, necessitating the development of new antimicrobial agents. Host defense peptides (HDPs) have been actively studied as promising antimicrobial agents due to their broad-spectrum activity and low susceptibility to resistance. However, natural HDPs are structurally unstable, leading to the exploration of HDP-mimicking peptides and amino acid polymers as synthetic antimicrobials. This review provides an overview of the design and applications of HDP-mimicking antimicrobial amino acid polymers, highlighting key structural characteristics.

The blood-brain barrier: structure, regulation, and drug delivery

Summary: The blood-brain barrier (BBB) is a natural protective membrane that prevents toxins and pathogens in the blood from entering the central nervous system (CNS). However, this barrier hinders the delivery of drugs to the brain, leading to low therapeutic efficacy and increased side effects. Recent advances in materials science and nanotechnology have provided advanced materials with customized structures and properties for targeted drug delivery. This review summarizes the physiological structure of the BBB and different cells contributing to its formation, as well as various strategies to enhance BBB crossing and regulate its permeability.

The easy synthesis and structural diversity of POXs suggest their potential as promising antifungal agents.

Secondary Amine Pendant β-Peptide Polymers Displaying Potent Antibacterial Activity and Promising Therapeutic Potential in Treating MRSA-Induced Wound Infections and Keratitis

Summary: Recent interest in developing antibacterial polymers as synthetic mimics of host defense peptides (HDPs) has focused on incorporating positively charged moieties to improve antibacterial activity and reduce eukaryotic toxicity. This study found that a secondary ammonium is superior to primary or tertiary ammoniums in antibacterial beta-peptide polymers, showing potent antibacterial activity and high selectivity. The optimal polymer displayed strong activity against antibiotic-resistant bacteria and showed high therapeutic efficacy with low toxicity.

Microbial Metabolite Inspired β-Peptide Polymers Displaying Potent and Selective Antifungal Activity

Summary: With the quick increase of serious invasive fungal infections and the limited antifungal drugs available, there is an urgent need for potent and selective antifungal agents. Inspired by microbial metabolites, the authors have designed and obtained poly(DL-diaminopropionic acid) (PDAP), a potent antifungal agent that kills fungal cells by penetrating the cytoplasm, generating reactive oxygen, and inducing fungal apoptosis. PDAP displays strong antifungal activity against Candida albicans, negligible hemolysis and cytotoxicity, and no susceptibility to antifungal resistance. In addition, PDAP effectively inhibits fungal biofilm formation and eradicates mature biofilms. In vivo studies have shown that PDAP is safe and effective in treating fungal keratitis, highlighting its potential as a promising new antifungal agent.

Blood-brain barrier-penetrating single CRISPR-Cas9 nanocapsules for effective and safe glioblastoma gene therapy

Summary: We designed a unique nanocapsule for efficient and safe gene therapy of glioblastoma. Our nanocapsules can penetrate the blood-brain barrier, target tumor cells, and selectively release the Cas9/sgRNA complex. Experiments demonstrated accurate and efficient gene editing in glioblastoma tissue with minimal off-target effects in high-risk tissues. Treatment with nanocapsules significantly extended survival time.

Discovery of Novel Orally Bioavailable Triazoles with Potent and Broad-Spectrum Antifungal Activity In Vitro and In Vivo

Summary: A series of novel triazole compounds containing 1,2,3-triazole side chains were synthesized and showed promising antifungal activity in vitro and in vivo. Compound 9A16 exhibited excellent antifungal and anti-drug-resistant fungal ability, as well as powerful efficacy in mice infected with various fungal strains. Furthermore, compound 9A16 demonstrated better anti-biofilm activity and resistance induction compared to fluconazole.

Small-molecule inhibitors for the Prp8 intein as antifungal agents

Summary: A small-molecule library was screened to find potent inhibitors of the Cne Prp8 intein, leading to the identification of compound 6G-318S with potential antifungal properties. These inhibitors showed specific action against Cryptococcus species and demonstrated synergistic effects with certain antimycotics. Treatment strategies for cryptococcal infection: challenges, advances and future outlook
