Induction of PARP7 Creates a Vulnerability for Growth Inhibition by RBN2397 in Prostate Cancer Cells

Chunsong Yang et al.

Summary: The ADP-ribosyltransferase PARP7 affects gene expression and protein function in prostate cancer cells by conjugating ADP-ribose to acceptor amino acids. A recently developed inhibitor, RBN2397, inhibits androgen-induced ADP-ribosylation in prostate cancer cells and reduces cell growth. RBN2397 also affects PARP7 localization and may be an actionable target in advanced prostate cancer.

Arginine ADP-Ribosylation: Chemical Synthesis of Post- Translationally Modified Ubiquitin Proteins

Jim Voorneveld et al.

Summary: This article describes the development and optimization of a methodology for preparing peptides and proteins modified on the arginine residue with an ADPr group. The authors successfully obtained four regioisomers of ADP-ribosylated ubiquitin using this method and investigated the activity and selectivity of Legionella pneumophila ligase and hydrolase enzymes through biochemical experiments.

Chemical genetics and proteome-wide site mapping reveal cysteine MARylation by PARP-7 on immune-relevant protein targets

Kelsie M. Rodriguez et al.

Summary: PARP-7 has been identified as an important regulator of the innate immune response, with a key target being the inactive PARP family member PARP-13. Specifically, PARP-7 preferentially MARylates cysteine residues in the RNA binding zinc finger domain of PARP-13, as revealed by proteome-wide ADP-ribosylation analysis.

Molecular Tools for the Study of ADP-Ribosylation: A Unified and Versatile Method to Synthesise Native Mono-ADP-Ribosylated Peptides

Jim Voorneveld et al.

Summary: ADP-ribosylation is crucial in DNA-repair, immunity, and other cellular processes with serine being the main acceptor. A new solid-phase strategy for synthesizing Ser-, Thr-, and Cys-MARylated peptides has been described. ADP-ribosylated cysteine is a stable bioisostere for ADP-ribosylated serine and the protein SpyMacroD from Streptococcus pyogenes acts as a Cys-(ADP-ribosyl) hydrolase.

Temporal and Site-Specific ADP-Ribosylation Dynamics upon Different Genotoxic Stresses

Sara C. Buch-Larsen et al.

Summary: The study identified early serine ADP-ribosylation events and observed an increase in modification of the same sites later on. Exposure to MMS-induced alkylating stress led to the strongest ADP-ribosylome response after 30 minutes, while H2O2-induced oxidative stress resulted in peak ADP-ribosylation after 60 minutes.

PARP7 mono-ADP-ribosylates the agonist conformation of the androgen receptor in the nucleus

Teddy Kamata et al.

Summary: The research uncovers the regulation of androgen receptor by PARP7 through ADP-ribosylation, which requires nuclear localization and agonist-induced conformation. The zinc finger structure of PARP7 plays a crucial role in modulating AR ADP-ribosylation, with effects separable from nuclear import.

Synthesis of ADP-Ribosylated Histones Reveals Site-Specific Impacts on Chromatin Structure and Function

Nir Hananya et al.

Summary: The study developed a efficient semi-synthesis method to create full-length ADP-ribosylated histones H3 and H2B, which were used to investigate the roles of histone ADP-ribosylation in DNA damage response. The results showed that ADP-ribosylation of serine-6 of histone H2B inhibits chromatin folding and higher-order organization, providing new insights into the impact of histone modifications on chromatin structure.
