Harnessing polymer-derived drug delivery systems for combating inflammatory bowel disease

Summary: The current clinical treatments for inflammatory bowel disease (IBD) suffer from severe side effects, low long-term treatment efficacy, and increased risks of infection and malignancy. However, naturally bioactive small molecules, reactive oxygen species scavengers, and gut microbiota modulators have emerged as promising candidates for IBD treatment. Polymeric systems have been engineered as delivery vehicles to improve the bioavailability and efficacy of these therapeutic agents, while reducing their systemic toxicity. This article surveys polymer-derived drug delivery systems for combating IBD.

Lysosomal nanotoxicity: Impact of nanomedicines on lysosomal function

Many nanomedicines have been withdrawn from post-surveillance due to safety issues. Lysosomal dysfunction caused by nanoparticles is emerging as the most common intracellular trigger of nanotoxicity. Physicochemical properties of nanoparticles have a great impact on their interaction with cells and subsequent toxicity. It is unjustifiable to generalize the safety and toxicity of nanoparticles as different particles possess distinct toxicological properties.

However, delivering drugs effectively to the eyes, especially for intraocular disorders, remains a major challenge due to various ocular barriers. Recent advancements in nanocarrier technology provide a promising solution by improving drug penetration, retention, solubility, toxicity reduction, release duration, and targeted drug delivery. This review highlights the progress and applications of nanocarriers, specifically polymer- and lipid-based nanocarriers, in treating different eye diseases and their potential for efficient ocular drug delivery. It also discusses ocular barriers, administration routes, and future prospects and challenges in the field of nanocarriers for ocular disease treatment.

Rapid identification and quantification of diquat in biological fluids within 30 s using a portable Raman spectrometer

Summary: In this study, a fast and simple detection strategy for diquat (DQ) in biological fluids was developed using surface-enhanced Raman spectroscopy (SERS). The method achieved point-of-care detection under acidic conditions with gold nanoparticles as the substrate. The strategy demonstrated high specificity and selectivity, and was validated using clinical methods. Compared to traditional clinical methods, SERS-based methods offer real-time measurements and have great potential for on-site and point-of-care testing.

This review examines the current understanding of starch structure, chemical modification methods, and their applications in drug delivery systems such as tablets, hydrogels, and patches. The information presented in this review can serve as a reference for future studies on chemically modified starch as excipients in drug carriers.

The surface coating of iron oxide nanoparticles drives their intracellular trafficking and degradation in endolysosomes differently depending on the cell type

Summary: This study investigated the intracellular transit of two magnetic nanoparticles (MNPs) with different surface coatings in mouse cell lines. The results showed that the coating type influenced the intracellular trafficking rate and the nature of the endolysosomes in macrophages. The MNPs with a specific coating had a slower transit rate and resulted in endolysosomes with more lytic enzymes and catalytic proteins in macrophages. These MNPs also induced more autophagic vesicles and enhanced the expression of iron metabolism-related genes and proteins.

A User-Friendly Platform for Single-Cell Raman Spectroscopy Analysis

Summary: The optimization of Raman instruments has expanded our understanding of single-cell Raman spectroscopy. By improving the speed and sensitivity, as well as implementing advanced data mining methods, complex chemical and biological information within Raman spectral data can be revealed. This paper introduces a new Matlab Graphical User-Friendly Interface (GUI) called CELL IMAGE, which enables the analysis of cellular Raman spectroscopy data. The GUI includes three main steps: spectral processing, pattern recognition, and model validation, and offers various well-known methods for each step. A new subsampling optimization method is integrated into the GUI to estimate the minimum number of spectral collection points. The incorporation of the signal-to-noise ratio (SNR) in the binomial statistical model makes the new subsampling model more sophisticated and suitable for complicated Raman cell data. These embedded methods allow CELL IMAGE to convert spectral information into biological information, including single-cell visualization, cell classification, and biomolecular/drug quantification.

Sustainable synthesis and theoretical studies of polyhedral gold nanoparticles displaying high SERS activity, NIR absorption, and cellular uptake

Summary: This study developed a simple method for preparing gold nanoparticles with multimodal properties. The nanoparticles were synthesized using trisodium citrate as a reducing, stabilizing, and shape-modulating agent at room temperature. The resulting gold nanoparticles had diverse shapes, good monodispersity, high surface-enhanced Raman scattering activity, and a wide absorption range. The optical properties and growth characteristics of the nanoparticles were investigated experimentally and numerically. In vitro studies demonstrated the biocompatibility of the nanoparticles with cells.
