Advances in cellular senescence in idiopathic pulmonary fibrosis (Review)

Shan Han et al.

Summary: Idiopathic pulmonary fibrosis (IPF) is a progressive, irreversible and fatal interstitial lung disease of unknown cause, with a median survival of 2-3 years. Aging may be linked to the development of IPF, but the mechanism remains unclear. Senescence of alveolar epithelial cells and lung fibroblasts (LFs) and their senescence-associated secretion phenotype (SASP) may play a role in IPF. This review focuses on the senescence of LFs and epithelial and stem cells, SASP, profibrotic signaling pathway activation, and potential treatments for IPF pathogenesis.

Hallmarks of aging: An expanding universe

Carlos Lopez-Otin et al.

Summary: Aging is driven by hallmarks that manifest with age, accelerate aging when accentuated experimentally, and can be decelerated, stopped, or reversed with therapeutic interventions. The twelve proposed hallmarks of aging include genomic instability, telomere attrition, epigenetic alterations, loss of proteostasis, disabled macroautophagy, deregulated nutrient-sensing, mitochondrial dysfunction, cellular senescence, stem cell exhaustion, altered intercellular communication, chronic inflammation, and dysbiosis. These hallmarks are interconnected with each other and with the recently proposed hallmarks of health.

Transcriptional changes of the aging lung

Minxue Jia et al.

Summary: This study provides a comprehensive and systemic analysis of the molecular signatures of lung aging using single-cell gene expression analysis. The research reveals that aging has a heterogeneous effect on lung cells, with some populations showing more transcriptional changes while others remain stable. The findings also highlight the importance of inflammation and immune response regulation in age-related lung pathologies.

Precision medicine advances in idiopathic pulmonary fibrosis

Theodoros Karampitsakos et al.

Summary: Idiopathic pulmonary fibrosis (IPF) is a highly heterogeneous and lethal chronic lung disease. Precision medicine approaches have not been effectively implemented in IPF clinical practice, despite the existence of numerous diagnostic, prognostic, and theragnostic biomarker candidates.

Semaphorin 3A in the Immune System: Twenty Years of Study

Ekaterina P. Kiseleva et al.

Summary: This article systematically analyzes the effects of Semaphorin 3A on T-lymphocytes, myeloid dendritic cells, and macrophages based on available literature from the past 20 years. The expression of Semaphorin 3A receptors in these cells is described in detail, and data from human and murine cells are compared. The article presents a comprehensive overview of the interaction between Semaphorin 3A and the mononuclear phagocyte system. Semaphorin 3A signaling mainly results in changes to the cytoskeletal machinery and cellular morphology, which regulate pathways involved in immune cell migration, adhesion, and cell-cell cooperation. It is increasingly recognized that Semaphorin 3A plays a crucial role in various phases of immune responses and is associated with many human diseases, making it a potential therapeutic target.

Mapping the immune environment in clear cell renal carcinoma by single-cell genomics

Nicholas Borcherding et al.

Summary: The study utilized single-cell RNA sequencing to analyze the tumor immune microenvironment of ccRCC patients and found an increase in CD8(+) T cells and macrophages in tumor-infiltrated immune cells, as well as identified a proliferation subpopulation associated with the progression of ccRCC. Utilizing gene expression analysis, the study also demonstrated the ability to predict clinical outcomes and pathological diseases based on subcluster assignment.
