Extracellular vesicles and nanoparticles: emerging complexities

Dennis K. Jeppesen et al.

Summary: The study of extracellular vesicles (EVs) and nanoparticles (NPs) is rapidly expanding, as recent discoveries have revealed a greater complexity and diversity than previously thought. New types of EVs and NPs have been described, showing that proteins and nucleic acids previously thought to be present in exosomes are actually more enriched in different types of EVs and in two recently identified amembranous NPs, exomeres and supermeres. This review provides an overview of the different types of EVs and NPs, highlights recent advances, and presents major unanswered questions.

A TLR4-independent critical role for CD14 in intracellular LPS sensing

Swathy O. Vasudevan et al.

Summary: CD14 plays a vital role in intracellular LPS sensing by mediating the cytosolic localization of LPS, leading to the activation of caspase-11. This study provides evidence for the critical role of CD14 in noncanonical inflammasome sensing of LPS.

Variation in blood microbial lipopolysaccharide (LPS) contributes to immune reconstitution in response to suppressive antiretroviral therapy in HIV

Zhenwu Luo et al.

Summary: This study found that bacterial lipopolysaccharide (LPS) enriched in the plasma of immune non-responders (INRs) in HIV patients contributes to poor CD4+ T cell recovery under suppressive ART. The LPS from different bacterial species have varying effects on CD4+ T cell apoptosis and proinflammatory responses.

NINJ1 mediates plasma membrane rupture during lytic cell death

Nobuhiko Kayagaki et al.

Summary: Plasma membrane rupture is a cataclysmic event in lytic cell death, releasing damage-associated molecular patterns to propagate the inflammatory response. NINJ1 protein plays an essential role in inducing PMR, with Ninj1-deficient macrophages showing impaired release of intracellular proteins and distinctive ballooned morphology. This study overturns the long-held belief that cell death-related PMR is a passive event, highlighting the active role of NINJ1 in mediating PMR.

Summary: Studies have identified galectin-1 as a new DAMP that promotes inflammation by inhibiting CD45. The release of galectin-1 during cytosolic LPS sensing is detrimental and affects endotoxin shock.

Hierarchical cell-type-specific functions of caspase-11 in LPS shock and antibacterial host defense

Puja Kumari et al.

Summary: The study reveals that macrophage/monocyte-specific caspase-11 plays a dominant role in mediating the pathological manifestations of endotoxemia, while caspase-11 expression in neutrophils is dispensable for LPS-induced lethality. Sensing of intracellular LPS by LyzM(+) myeloid cells and MRP8(+) neutrophils is necessary for bacterial clearance and host survival during intracellular bacterial infection.

Quantitative proteomics identifies the core proteome of exosomes with syntenin-1 as the highest abundant protein and a putative universal biomarker

Fernanda G. Kugeratski et al.

Summary: The study used an unbiased and quantitative proteomic approach to identify core constituents of exosomes, with syntenin-1 identified as a potential universal biomarker. A comprehensive quantitative atlas of core proteins ubiquitous to exosomes was provided, highlighting the potential use of syntenin-1 as a putative universal biomarker for exosomes.

Inflammasome-induced extracellular vesicles harbour distinct RNA signatures and alter bystander macrophage responses

Christina F. Budden et al.

Summary: The text explains that activation of inflammasomes can lead to inflammation and immune pathology, but EVs released by inflammasome-activated cells also contain specific RNA, which can induce interferon beta in bystander cells to suppress NLRP3 inflammasome responses.
