Optical fiber amplifier and thermometer assisted point-of-care biosensor for detection of cancerous exosomes

Summary: A colorimetric and photothermal point-of-care testing (POCT) biosensor based on rolling circle amplification (RCA) was developed for sensitive detection of cancerous exosomes. The biosensor showed colorimetric and photothermal responses, which were recorded using an optical fiber amplifier and a forehead thermometer, respectively, enabling hand-held detection. The biosensor exhibited excellent specificity and resistance to environmental interference.

Controllable color change of polydiacetylene vesicles under thermal-photo stimuli

Summary: Polydiacetylene (PDA) exhibits unique color transition properties in response to external stimuli, making it a promising material for optical biosensors. However, the irreversible color transitions of most PDA materials limit their applications. This study demonstrates that the colorimetric reversibility of PDA is closely related to the difference in hydrogen bond strength and ionic interactions between adjacent headgroups of PDA vesicles. By comparing the reversibility of PDA vesicles with different terminal functional groups, it was found that specific compositions of PDA vesicles show noticeable color reversibility during thermal-photo stimuli cycles.

Electrochemical micro-aptasensors for exosome detection based on hybridization chain reaction amplification

Summary: This study presents an electrochemical micro-aptasensor for highly sensitive detection of exosomes, integrating micropatterned electrodes with a hybridization chain reaction (HCR) signal amplification method. The sensor enables specific detection of cancerous exosomes in serum samples with a wide linear detection range and high sensitivity. The method shows great potential for early cancer diagnosis by successfully detecting lung cancer exosomes in both early-stage and late-stage lung cancer patients' serum samples.
