Nrf2 Activation in Chronic Kidney Disease: Promises and Pitfalls

Ana Karina Aranda-Rivera et al.

Summary: The nuclear factor erythroid 2-related factor 2 (Nrf2) plays a role in protecting cells against oxidative damage and is linked to the progression of chronic kidney disease (CKD) and CKD-related morbidity. The activity of the Nrf2 system is influenced by factors such as inflammation, kidney fibrosis, mitochondrial and metabolic effects, and the accumulation of uremic toxins and inflammation. The activation or repression of Nrf2 varies depending on the stage and cause of kidney disease.

Clinical Improvement of Disseminated Acanthamoeba Infection in a Patient with Advanced HIV Using a Non-Miltefosine-Based Treatment Regimen in a Low-Resource Setting

Denasha L. Reddy et al.

Summary: Disseminated Acanthamoeba infection is often overlooked and underdiagnosed in HIV patients in South Africa. The difficulty in diagnosis and limited management options in low-resource settings pose unique challenges. This case highlights the need to consider Acanthamoeba infection in advanced HIV patients with nodular and ulcerative skin lesions, and emphasizes the importance of advocating for better access to miltefosine in South Africa for the treatment of acanthamoebiasis.

Histological Changes in the Kidneys and Heart in Experimental Acanthamoebiasis in Immunocompetent and Immunosuppressed Hosts

Karolina Kot et al.

Summary: The course of Acanthamoeba spp. infection varies depending on host age and immune status, as well as strain virulence. Histopathological changes were observed in the kidneys and heart of immunocompetent and immunosuppressed mice infected with Acanthamoeba spp., indicating the importance of analyzing gene and protein expressions to better understand infection progression in these organs.

Leishmania donovani Metacyclic Promastigotes Impair Phagosome Properties in Inflammatory Monocytes

Christine Matte et al.

Summary: Leishmaniasis, caused by protozoan parasites, can have debilitating effects ranging from self-healing ulcers to life-threatening visceral pathologies. Research has shown that early-stage interactions between Leishmania and inflammatory monocytes can impact antimicrobial responses, with the parasites inhibiting microbicidal properties of phagosomes within the cells. This study found that Leishmania donovani metacyclic promastigotes can impair superoxide production and pH acidification within inflammatory monocytes, potentially contributing to the establishment of chronic disease.

The antioxidant response favors Leishmania parasites survival, limits inflammation and reprograms the host cell metabolism

Marta Reverte et al.

Summary: The activation of NRF2 is crucial in protecting Leishmania parasites from oxidative stress during infection. NRF2 is strongly upregulated in Leishmania infection and plays a role in parasite survival. It is dependent on NOX2 and SFKs signaling pathways and reprograms host cell metabolism, showing a dual role in promoting parasite persistence and limiting inflammation.

Potential Biomarkers in Diagnosis of Renal Acanthamoebiasis

Karolina Kot et al.

Summary: Recent studies have shown that Acanthamoeba spp. may contribute to kidney dysfunction. The levels of KIM-1, NGAL, and MCP-1, as well as the activity of MMP-2 and MMP-9, were examined in the kidneys of immunocompetent and immunosuppressed mice infected with Acanthamoeba spp. The results suggest that KIM-1, NGAL, MCP-1, MMP-2, MMP-9, and MMP-9/NGAL could be potential biomarkers for renal acanthamoebiasis.
