Hypermethylation of RNF125 promotes autophagy-induced oxidative stress in asthma by increasing HMGB1 stability

Summary: This study found that RNF125 gene expression was downregulated in the bronchial epithelium of asthma patients and mice, and this low expression was caused by hypermethylation of the Rnf125 gene. Furthermore, it was found that RNF125 could inhibit autophagy and oxidative stress, and protect the epithelial barrier. This study revealed that the hypermethylation of the Rnf125 gene reduced its expression, thereby promoting autophagy-induced oxidative stress in asthma by increasing HMGB1 stability.

Summary: Hepatocellular carcinoma (HCC) is a leading cause of cancer-related deaths worldwide, but its tremendous inter- and intra-tumor heterogeneity has made it difficult to identify driver genes for HCC. By analyzing transposon mutagenesis screening data in the liver, researchers have discovered a new tumor suppressor gene, RNF125. This gene functions as a negative regulator of cell proliferation through transcriptional suppression of multiple genes important for cell proliferation and liver regeneration. This gene is frequently inactivated in human HCC and has a significant impact on patient prognosis.
