The role of platelets in immune-mediated inflammatory diseases

Summary: Immune-mediated inflammatory diseases (IMIDs) are characterized by excessive and uncontrolled inflammation and thrombosis, both of which are responsible for organ damage, morbidity and death. Platelets play a central role in the pathogenesis and progression of IMIDs, as they are activated by disease-specific factors and promote inflammation and tissue injury. Targeting platelet activation and their interaction with the immune system are promising therapeutic strategies in IMIDs.

This review focuses on the structural-functional domains of CCR7 and its impact on dendritic cell migration to lymph nodes, as well as the regulatory network of CCR7. Furthermore, potential strategies targeting the CCR7 network to regulate dendritic cell migration and manage inflammatory diseases are discussed.

Liposome induction of CD8+ T cell responses depends on CD169+ macrophages and Batf3-dependent dendritic cells and is enhanced by GM3 inclusion

Summary: In this study, liposomes containing GM3 were shown to efficiently target CD169(+) macrophages, inducing specific immune responses including antigen-specific CD8(+) and CD4(+) T cell as well as B cell responses. Administration of GM3 liposomes containing ovalbumin resulted in delayed tumor growth and improved survival in B16-OVA tumor bearing mice. The study also highlighted the essential role of CD169(+) macrophages and cDC1s in inducing CD8(+) T cell immunity through liposomal vaccination.
