Catalytic Asymmetric Synthesis of Axially Chiral Diaryl Ethers through Enantioselective Desymmetrization

Summary: In this study, a highly efficient catalytic asymmetric synthesis of diaryl ether atropisomers was reported via an organocatalyzed enantioselective desymmetrization protocol. The chiral phosphoric acid-catalyzed asymmetric electrophilic aromatic aminations of symmetrical substrates provided a series of diaryl ether atropisomers with excellent yields and enantioselectivities. The utilization of the 1,2-benzenediamine moiety in the products allowed for the facile construction of heterocycles, leading to a wide range of structurally diverse and novel azaarene-containing diaryl ether atropisomers.

Atroposelective Silylation of 1,1′-Biaryl-2,6-diols by a Chiral Counteranion Directed Desymmetrization Enhanced by a Subsequent Kinetic Resolution

Summary: A desymmetrizing silylation of aromatic diols is achieved by using List's counteranion directed silylation technique for asymmetric silyl ether formation of phenol derivatives. The enantioselective discrimination of achiral 1,1'-biaryl-2,6-diols is enabled by a silylium-ion-like silicon electrophile generated from an allylic silane paired with an imidodiphosphorimidate (IDPi). The enantioselectivity of the desymmetrization is further improved by a downstream kinetic resolution, converting the monosilylated minor enantiomer into the corresponding bissilylated diol.

N-Heterocyclic Carbene-Catalyzed Atroposelective Synthesis of N-N Axially Chiral 3-Amino Quinazolinones

Summary: While the selective synthesis of biaryls with axially chiral C-C bonds is well-known, the synthesis of compounds containing axially chiral C-N bonds is less explored, and the construction of axially chiral N-N bonds has received little attention. This study demonstrates the selective amidation reaction catalyzed by N-heterocyclic carbenes (NHCs), leading to the atroposelective synthesis of N-N axially chiral 3-amino quinazolinones. The NHC-catalyzed reaction of quinazolinones with a free N-H group and α,β-unsaturated aldehydes under oxidative conditions provides the atropisomeric quinazolinone derivatives under mild conditions and with a broad scope. Preliminary studies on experimental and density functional theory-based determination of N-N rotational barrier are also presented.

A Dynamic Kinetic Resolution Approach to Axially Chiral Diaryl Ethers by Catalytic Atroposelective Transfer Hydrogenation

Summary: This paper presents a dynamic kinetic resolution method for the synthesis of axially chiral diaryl ethers. By using a Bronsted acid catalyzed atroposelective transfer hydrogenation (ATH) reaction, the desired diaryl ethers could be obtained in moderate to good chemical yields (up to 79%) and high enantioselectivities (up to 95% ee) under standard reaction conditions. These structural motifs are interesting precursors for further transformations and may have potential applications in the synthesis of chiral ligands or catalysts.

N-Heterocyclic Carbene (NHC)-Catalyzed Transformations Involving Azolium Enolates

Summary: This article highlights the recent advances in the N-heterocyclic carbene (NHC)-organocatalyzed generation of azolium enolate intermediates and their subsequent interception with electrophiles. The NHC-bound azolium intermediates are generated by adding NHCs to suitably substituted aldehydes, acid derivatives, or ketenes. A wide range of coupling partners can intercept the azolium enolates to form [2+n] cycloadducts (n=2,3,4) and various alpha-functionalized compounds. Enantioselective synthesis of the target compounds is achieved with chiral NHCs.

Synthesis, structures, and photophysical properties of three-coordinate copper(I) complexes bearing bidentate bis[(2-diphenylphosphino)phenyl]ether (POP) ligand and monodentate substituted-quinoline ligand

Summary: In this study, three-coordinate Cu(I) complexes were synthesized and characterized. The photophysical properties of these complexes were tuned by introducing organic groups into the quino-line ligand, resulting in a lower energy metal-to-ligand charge transfer band. The emissions in solid state exhibited wavelengths in the range of 542-558 nm, with high quantum yields and long-lived excited lifetimes. The photophysical properties were further explained using DFT and TDDFT methods.

Summary: The first atroposelective synthesis of pyrrolo[3,4-b]pyridines catalyzed by N-heterocyclic carbene has been achieved, leading to a wide range of chiral atropisomers with high yields and excellent enantioselectivities (96-99% enantiomeric excess). Experimental results and density functional theory calculations demonstrate the high thermal stability of the C-N axial chirality in the product.

An Efficient and Sustainable Synthesis of the Antimalarial Drug Tafenoquine
