B(C6F5)3-Catalyzed Diastereoselective and Divergent Reactions of Vinyldiazo Esters with Nitrones: Synthesis of Highly Functionalized Diazo Compounds

Summary: Here, a mild, transition-metal-free, highly diastereoselective Lewis acid catalyzed methodology for the synthesis of isoxazolidine-based diazo compounds from the reaction between vinyldiazo esters and nitrones is reported. Interestingly, the diastereoselectivity of the isoxazolidine products is found to be contrasting to previous metal-catalyzed reactions. Furthermore, the same catalyst can be used with enol diazo esters, leading to the formation of Mukaiyama-Mannich products. These diazo products can be further functionalized to generate benzo[b]azepine and pyrrolidinone derivatives.

Rh(III)-Catalyzed C-H Functionalization/Annulation of 1-Arylindazolones: Divergent Synthesis of Fused Indazolones and Allyl Indazolones

Summary: Rh(III)-catalyzed C-H/N-H annulation and C-H allylation of phenylindazolones have been achieved using 5-methylene-1,3-dioxan-2-one and 4-vinyl-1,3-dioxolan-2-one as scalable cross-coupling partners, providing functionalized indazolone fused heterocycles and branched and linear allyl indazolones with moderate to high yields. These synthetic protocols demonstrate mild conditions, broad substrate scope, and high functional-group compatibility. Additionally, scale-up synthesis and preliminary mechanistic exploration were also conducted.

Rh(III)-Catalyzed Spiroannulation Reaction of N-Aryl Nitrones with 2-Diazo-1,3-indandiones: Synthesis of Spirocyclic Indole-N-oxides and Their 1,3-Dipolar Cycloaddition with Maleimides

Summary: An efficient strategy for the preparation of spirocyclic indole-N-oxide compounds through a Rh(III)-catalyzed [4 + 1] spiroannulation reaction of N-aryl nitrones with 2-diazo-1,3-indandiones as C1 synthons under extremely mild conditions is presented. From this reaction, 40 spirocyclic indole-N-oxides were easily obtained in up to 98% yield. In addition, the title compounds could be successfully used for the construction of structurally intriguing maleimide-containing fused polycyclic scaffolds via a diastereoselective 1,3-dipolar cycloaddition reaction with maleimides.

One-Pot Synthesis of Benzo[c]phenanthridine Alkaloids from 7-Azabenzonorbornadienes and Aryl Nitrones

Summary: An efficient synthesis of benzo-[c]-phenanthridine alkaloids via a synergistic combination of C-C bond formation and a cycloaromatization reaction is described. Aryl nitrones react with 7-azabenzonorbornadienes in the presence of a Rh-(III) catalyst, providing pharmaceutically useful benzo-[c]-phenanthridine derivatives in good to moderate yields. Using this methodology, highly useful alkaloids such as norfagaronine, norchelerythrine, decarine, norsanguinarine, and nornitidine were prepared in a single step.

Additionally, the achievement in the synthesis of high-oxidation state Securinega alkaloids is depicted, including oxidative transformations and fragment-coupling strategies. These methods and strategies have broad applicability to other alkaloids and can expand the chemical space of secondary metabolites.

The use of N-functionalized cyclic carbonates is crucial for efficient catalytic turnover, leading to the preparation of a broad range of amino-isocoumarin derivatives with satisfactory yields. One derivative, an amino-isocoumarin conjugated with a selenocyano functionality, was found to be nearly four times as active as the marketed drug abiraterone against T47D cancer cells.
